This paper is aim at the study of the isolation Lactarius deliciosus hypha,growth conditions of Lactarius deliciosus,extraction mycelium polysaccharide and its'pharmacological activity preliminary.Work and specific conclusions are as follows:(1)Study on the Strain Breeding and Growth Conditions of Lactarius deliciosus hypha.Eight strain of Lactarius deliciosus were separated from different parts of Lactarius deliciosus.The results showed that the mycelia separated from petiole grew fastest,the optimal carbon source was glucose and the optimal nitrogen source was Ammonium acetate followed by the corn pulp and peptone.Furthermore,the optimal PH of the mycelia growth was different under different media.(2)Three methods extraction of Lactarius deliciosus polysaccharides at intracellular①The optimum extraction polysaccharide process of microwave technology was gained by using a single-factor test.The results showed that the optimal extraction conditions as the following: extraction time is 120 S~180S,The proportion of materials and water is 1.0 to 2000~3000and the alcoholic content is 15%to 25%.②The optimum extraction polysaccharide process of complex emzyne technology were determine by using a single-factor test.The results showed that the optimal extraction conditions as the following:extraction PH value was 5.0,extraction temperature was 50℃,The proportion of materials,water and complex emzyne was 100:50000:1.5(1∶2)③Through experiments and single factor orthogonai experimental to determine the optimum extraction polysaccharide process of Ultrasonic extraction.The results showed that the optimal extraction as the following conditions,ultrasound 60 minutes,ultrasonic power to 400 W, mycelium Size of screen for over 60 heads,plus water,250 ml,ultrasound temperature of 60℃.(3)Study on Pharmacological Activity of Lactarius deliciosus hypba,polysaccharides①The bacteriostasis in vitro of intracellular polysaccharides and extracellular polysaccharides of mycelium in Lactarius deliciosus on five common pathogenic bacterias,E.coli,salmonella, staphylococcus,streptococcus,Pasteurell,were determined,and the minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) of the two polysaccharides against the five pathogenic bacterias above were gained.②The effect of intracellular polysaccharides and extracellular polysaccharides in Lactarius deliciosus mycelium against IBD Virus and ND Virus were observed.It is concluded that extracellular polysaccharides of mycelium in Lactarius deliciosus have no effect on IBD Virus and ND Virus,whereas intracellular polysaccharides of mycelium in Lactarius deliciosus have hacteriostasis on IBD Virus at a certain extent.
