Pomelo which is a very important fruit crop, has long history. A common problem, which limits identification and utilizing, is the homonym and synonym caused by pomelo resources exchanging. Lots of works by using morphology, pollenand isoenzyme and growth chafer once focused on identification, taxonomy and evolution of pomelo. But there are still some divergence and limitation. Here, genetic diversity of 31 pomelo materials of our province had been conducted by using RAPD. The main results are as follows:1 .Based on the analysis of three pomelo genomic DNA isolated methods. The improving SDS method is best one. Tissue extraction with SDS in the presence of a high salt concentration suppressed co-precipitation of polysaccharides and DNA. DNA preparations were free of gelatinous substances.2.Some essential factors that might affect the result of RAPD assay were tested and compared. An optional reacti'on system suitable for the assay and usage of RAPD in taro was established. The volume of amplified reaction was 25ul,containing 10× buffer (Mg2+, 20mM) 2.5ul, genomic DNA (40ng/ul) 0.5ul, primer(5uM) 2ul, dNTP (10mM) 0.25ul, Taq DNA ployrase (2U/ul) 0.5ul, and pure water 19.25ul. The reaction program was devised for 42 cycles, each with 93℃ denaturation for lmin, 36℃ annealing for lmin, 72℃ extension for 2min, and93℃ predenature for 2min in the former denature and 72 ℃ postextension for 10min in the final extension.3.Thirty that were selected out from one hundred decameter arbitrary primers,had polymorphisms and dear bands, and then were successfully amplified in analysis of pomelo germplasm.4. When genetic distance is 0.24, all of materials are classified to 4 categories. Besides every kind has a specific band.
