Tomato is self-pollination plant, and the heterosis was obvious. In contrast to the cultivar, tomato hybrid increased by 20-30% in yield, with higher regularity and better resistance . So the investigation to tomato male sterility plays an important role in tomato hybrid breeding.We introduced the tomato CMC line with ps-2 male sterility gene from Bulgaria, and study on CMC1ps2 line which was chosed from the CMC line . We found out CMC1ps2 line grew very well in Beijing, and the character of male sterility was normal. The 100% sterility infant easily were got through selfing, and the fertility F1 were obtain which derived from CMC1ps2 as a male parent and a common tomato as a female parent . So CMC1ps2 line has been conveniently used for hybrid breeding.In this study, 123 individuals of F2 segregated population were derived from Bulgaria line CMC1ps2 as a male parent and a virtual processing tomato line 92155 as a female parent. The segregation ration of fertility and sterility in this population completely meet the Madeline genetic mechanism as a single recessive gene. AFLP ,SSR,and CAPS technology have been used to obtain the markers tightly liked with ps-2 gene. 64 AFLP ,5 SSR and 12 CAPS primer pairs were screened and 3 AFLP markers including E37/M47 , E38/M48 and E33/M50 , a SSR marker SSR450 and a CAPS marker PStgl23 have been identified to tightly linked with ps-2 loci. The genetic distances were respectively 6.04 cM, 6.04 cM, 6.06 cM, 4.937 cM and 11.591 cM. The 3 AFLP markers and the SSR marke were located one side with ps-2 loci and all the position was almost same.The CAPS marker PStg123 were located anther side with PS-2 loci. The 3 AFLP markers were dominant and linkde with ps-2 loci. The SSR marker and the CAPS marker were codominance. These 5 markers obtained in this study couid be directly used for maker-assistant selection and speed up the introgression and use of male sterility for tomato production.And the DNA fragment amplified with E37/M47 marker has been retrieved and sequenced in order to transform AFLP marker into SCAR marker, and a sequence of 392 bp was obtained. These works were a base to some futher study about the sequence of ps-2 gene, the gene cloning and the transgenic vegetable etc.
