Tomato bacterial speck is a severe worldwide disease that affects the production and quality of tomato. With the disease-resistant materials of LA3472,BL984,Huang zhenzhu and susceptible materials of R,H, the study designed specific primers according to resistance gene Pto that published in Gene Bank by Martain, and the genome DNA of all materials were amplified by PCR. The restriction enzyme analysis was based on the PCR amplification fragments. In study, the CAPS labeled molecular technique system has been established for disease-resistant materials of tomato bacterial speck. The main content as follows: Specific primers design for disease resistance genes; Establishment of PCR reaction system; Restriction endonuclease selection; Restriction fragment length polymorphism detection.;screen out the CAPS marker of pto gene, PCR(pto7) products with 550bp were digested by RsaI,which is restriction fragment length polymorphism detection.These results had very important significance of molecular breeding about tomato bacterial speck .
