| Madicago sativa L. is a protein -rich perennial leguminous plant and the crude protein content of dry matter is always around 20%. A large number of researches have proved that Madicago sativa L. is one of the fittest plants for leaf protein extraction. Based on the material of Madicago sativa L. in southern areas, through optimizing the method of leaf protein extraction, and comparing extraction effects of ten different methods, a series of leaf protein extraction methods respectively fitting for the needs of lab research and large-scale production of factories was proposed. In addition, effect of wiping off grass smell on alfalfa leaf protein was conducted by fermenting methods; the experiment provides a wider applicable method for alfalfa leaf protein.The results indicated that:1.The optimal technique for lab extracting the leaf protein of Madicago sativa L.: by optimizing the leaf protein extraction method in various aspects, such as water bath temperature and time, alfalfa mincing length, the water amount and spending time in breaking up process, centrifugal rev and time, the best technique parameter of lab leaf protein extraction method was got that water bath temperature was 90 ℃, water bath time 10min, mincing length 2cm, the proportion between water and alfalfa 1: 1.2, breaking up time 2min, centrifugal rev 3000rmp/min and centrifugal time was 15min.2.The optimal method of alfalfa leaf protein extraction: compared the results of following ten extraction methods: direct centrifuge, salting out treatment, organic impregnant treatment, flocculant treatment, heating up treatment, acidification treatment, basification treatment, acidification heating up treatment, basification heating up treatment and ferment treatment, it was clear that acidification treatment and flocculant treatment were two promising methods. The result of leaf protein extraction by PH5 treatment was shown as: the amount of extracted material being 1.82g, the content of coarse protein in extracted material being 43.99%, the total amount of coarse protei being 0.80g, respectively increasing 157%, 13.1%, 281% compared to direct centrifuge, and the result by 5% aluminium potassium sulfate was that the amount of extracted material was 1.77g, the content of coarse protein 38.90% , the total amount of coarse protein 0.69g, respectively increasing 234%, -0.03% , 229% compared to direct centrifuge.3.The most efficient combination treatments for dipping off grass smell through ferment: Experimenting on such matters as EM, fermentative acid, sucrose and Etuoxyquin dry powder, the result indicated that all of them were capable of wiping off the smell, and with their amount increasing in the process, the amount of smell residue decreased. Among these four factors, EM and fermentative acid were most effective. There existed a significance interaction among the four factors in the EM treatment.
