Wheat Genetic engineering provides a complementary tool to conventionalbreeding, and increases in genetic variability in wheat germplasm by the introduction of genes through genetic transformation. Transformation for elite wheat cultivar with good agronomic traits can shorten breeding cycle and provide unique genetic materials.The present study utilized mature seeds of 11 different wheat genetypes as the starting material cultured in vitro, and four genetypes with good agronomic characteristics were selected out, which displayed a high regeneration response. Furthermore, a high frequency of plant regeneration system from mature embryos of the four elite wheat cultivars was established. On basis of this, mature embryo-mediate calli of wheat (Triticum aestivum L.)cultivars chuannongl2 chuannongl7 chuannong18 and mianyang11 were bombardmented using PDS 100/He particle delivery system in the presence of plasmid pBI 121,which contained a cycd2 gene and selectable marker NptII gene .PCR and Southern blot revealed the integration of transgenes in the TO transformants., and four plants were identified to be the transgenic wheats containing the target genes. The general result is following:1. Mean callus induction rate of eleven genetypes was over 90%, but plant regeneration rate of callus initiated from mature embryo was more than 45% in four cultivars wheat, less than 26% in seven wheat lines. Therefore the four wheat cultivars were good for culture in vitro.2. Intact mature embryos and half-mature embryos(lack of radicel) of eleven wheat genetypes were cultured in vitro .The results indicate that half-mature embryos culture can decrease efficiently germination of embryos in callus induction media.Therefore...
