As one common diseases of the nervous system, Stroke is a serious threat to human health and life, In addition to traditional risk factors such as high blood sugar, high blood pressure, high cholesterol, smoking, and other related, the genetic variation can do impact to stroke, environmental and genetic factors can do effection on ischemic stroke respectively, also they can increase the susceptibility of stroke through the interaction of them. Leukotriene A4hydrolase (LTA4) is the key enzyme in the process of leukotriene generation, many researches about its genetic polymorphisms focus on cancer and respiratory system disease, there are few studies to explore its relationship with ischemic cerebral apoplexy, this study aims to serve LTA4H as candidate genes to investigate its effection of genotype polymorphism on atherosclerotic cerebral infarction and provide clinical data for the individual prevention and treatment of this disease.Objective:To explore the association between polymorphism of leukotriene A4hydrolase gene on atherosclerotic cerebral infarction(ACI).Methods:This experiment regard Shandong population as the research object to explore the association between polymorphism of leukotriene A4hydrolase gene on atherosclerotic cerebral infarction by the case-control study and molecular epidemiological studies method, The case group consisted of450patients with artery atherosclerosis ischemic stroke recruited from the Affiliated Hospital of Qingdao University Medical College in China during October2010to October2012. Diagnostic criteria of ischemic stroke in accordance with criteria of the Chinese Medical Association in1995and the criteria amended in the fourth national cerebrovascular disease conference and Trial of Org10172in Acute Stroke Treatment (TOAST) maded by Adams in1993. In addition to clinical diagnosis, each patient was examined by neck vascular ultrasound and magnetic resonance angiography (MRA) as well as computerized tomography angiography(CTA) or digital subtraction angiography(DSA) to confirm the diagnosis. The control group included400individuals recruited from neurology department, other departments and health physical examination center Controls were sampled to match stroke patients for age and gender. One SNP (rs6538697) of theLTA4H gene was genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method to test the association between the genetic factors and atherosclerotic cerebral infarction.Results:The There were significant differences between patients and healthy subjects in the genotype distribution of LTA4H(X2=4.49, P<0.05), the C allele frequency in atherosclerotic cerebral infarction group was significantly higher than that in the control group, the difference was significant and the T allele frequency in ischemic stroke group was lower than that in the control group(X2=7.27, P<0.05), Logistic regression analysis were used to get rid of traditional risk factors as Hypertension, Diabetes, Triglyceride(TG),Total cholesterol(TC),Low density lipoprotein cholesterol(LDL).For LTA4H rs6538697, Carriers of C allele in rs6538697were more susceptible to atherosclerotic cerebral infarction (OR=1.34,95%CI1.010-1.786, P<0.05).Conclusion:There is a correlation between LTA4Hrs6538697C/T gene polymorphism and the atherosclerotic cerebral infarction in Shandong population, rs6538697T→C allelic gene mutations can increase the risk of atherosclerotic cerebral infarction.
