Objective:Atherosclerosis (AS) is the pathologic foundation in the formation and development of brain vessel disease. Oxidative stress (OS) participates the entire process of AS, which plays a key role in the ischemic stroke. GSTs (glutathione S-transferase) together with other antioxidant enzymes eliminate the active oxygen species (ROS) to protect cells against the oxidative damage. Currently it is not well known whether the polymorphisms of glutathione isozyme (GSTT1, GSTM1) exist in Chinese ischemic stroke patients. The present study aims to assess the association of glutathione isozyme (GSTT1, GSTM1) gene polymorphisms with ischemic stroke risk in the Chinese Han Population, and discuss the mechanism of action in ischemic cerebral vascular diseases.Method:We selected315ischemic stroke patients and227physical examination persons to establish a case control study model, determined the GSTT1, GSTM1genotype via the polymerase chain reaction (PCR), gel electrophoresis, gene sequencing methods and followed by analyzing the data using statistic software.Result:(1) No obvious evidence of GSTT1null, GSTM1null and GSTT1/GSTM1double null genotype distribution differences between the case group and control group was found, the same result was obtained in the sex subgroups.(2) The stroke risk increased apparently in the following situation:hypertension accompanied by GSTT1null genotype (OR=2.996,P<0.001), hypertension accompanied by GSTM1null genotype (OR=3.680, P<0.001), DM (diabetes mellitus) with GSTT1null genotype (OR=1.860,P=0.031), DM with GSTM1null genotype (OR=2.444,P=0.002), smokers with GSTT1null genotype (OR=2.276, P=0.003)Conclusion:GSTT1null genotype, GSTM1null genotype may interact synergistically with hypertension, diabetes or smoking to increase the incidence risk of ischemic stroke.
