Objective:The methods including thin-layer chromatography,high performance liquid chromatography (HPLC), ultra high performance liquid chromatography-mass spectrometry (UPLC-MS) had been established for nine synthetic pigments, which probably contained Trartrazine, Amaranth, Ponceau4RC, Acid Violet7, Allura Red AC, Brillant Yellow, Solvent Yellow34, Orange2sodium salt, Erythrosin B dinodium salt in four medicine of Pollen Typhae, Lycium barbarum, red flower, baicalin and capsule. Methods:The developing solvent based on ethyl acetate:anhydrous ethyl alcohol:water:aqueous ammonia (6:4:2:0.5) had been used in thin-layer chromatography to quickly screen nine synthetic colouring matter in four medicine of Pollen Typhae, Lycium barbarum, red flower, baicalin and capsule. The contents had been detected by HPLC based on the optimum conditions, the methanol-50mmol/L ammonium acetate had been used as mobile phase for gradient elution, the wavelengths for measurement were405nm、435nm、485nm、520nm, the flow rate is1.0mL/min, the column temperature is35℃and used10μL as sample amount for loading. The finally validation was been tested by UPLC-MS for nine coloring matter in medicine.Resluts:The isolation spots of nine synthetic pigments had been displayed in visible light after the development of samples, the same color spots and Rf values had been detected for positive sample and reference substance. The good linear range was4~18μg/mL correlation coefficient was0.9999, the recovery with standard additional was90.81%~100.34%, RSD was lower than3%. The retention time, molecular weight of the parent ion and fragment ion were in accordance with possible synthetic pigments in medicine and capsule by using LC-MS method. Conclusion:The three methods had been applied for the qualitative and quantitative determination of the illegally used nine synthetic pigments in medicine.
