OBJECTIVE: Illegally dyeing Chinese herbs exist in the domestic. Substandard medicines are poisonous, and they will bungle diseases. In order to establish different methods of qualitative analysis and quantitative determination of the pigments by HPTLC、HPLC、UPLC-MS/MS.METHODS:The pigments are divided into 3 categories: red, yellow and dark. Using the following 3 kinds of methods analyze them: Qualitatively screen the positive samples and the pigments by TLC and HPTLC simultaneously and determine the environmental effects on experiment of water-soluble pigments. The 31 contents were determined by HPLC with three-wavelength at the same time. Verification results by UPLC-MS/MS.RESULTS: Detect Abietic acid after treating sample, then detect other water-soluble pigments and fat-soluble pigments by TLC and HPTLC simultaneously. All the pigments could completely separated with 420 nm, 500 nm and 600 nm wavelength and exhibited good linearity within the concentration range of 0.19 to 27.3 μg/m L, the lowest detection limit was 0.01 μg/m L. Mass spectral information of the pigments are detected and the UPLC-MS/MS results are consistent with the previous results.CONCLUSION: We establish 3 kinds of methods of qualitative analysis and quantitative determination of the pigments by TLC,HPLC and UPLC-MS/MS. The methods can carry out the qualitative and quantitative analysis with high accuracy, sensitivity and simplicity, it is suitable for the screening and identification of the pigments illegally added to Chinese herbs. |
