Uyglur herb snow chrysanthemum is the dry flower heads of Coreopsis tinctoria Nutt.. In recent years, it is well known and widely used for preventing and treating the coronary heart disease (CHD). Modern pharmacological research revealed that the flavonoids in snow chrysanthemum can inhibite the proliferation and differentiation of 3T3-L1 preadipocytes and can prevent the rising of total lipid and improve lipid metabolism in hyperlipidemia mice to show a good effect in descending blood-fat. Howere, the main compounds of the flavonoids are remaining unknown. It is inherent urgency to carry out this research. So, this paper research on the enrichment process and chemical characterization of the flavonoids in snow chrysanthemum.1. The extraction process of snow chrysanthemum and the enrichment process of the flavonoids were studied and optimized with mono-factorial experiments and orthogonal design.The optimum extraction conditions were as follows:Smashed the crude herb to screen through mesh size of 24, then extracted with percolation method. Wetted 2 hours with 2 times of 50% methanol. And then transferred to a diacolation device and soaked for 24h with 40 times of 50% methanol before diacolation. The percolate rate was 3mL/min.The optimal enrichment conditions were as follows:AB-8 macroporous resins were used for enriching the flavonoids in snow chrysanthemum. The largest sample amount was 3.5BV of extracts of snow chrysanthemum (0.0987g/ml). The eluting solvent was 60% methanol, the ratio of diameter to height was 1:10 and the percolate rate was 3BV/h.2. To separate and purify the flanonoid compounds by column chromatography and semi-preparative HPLC. Finally, marein and flavanomarein have been obtained and the structures of them have been elucidated by 1H-NMR and 13C-NMR.3. At last, this study employs the technology of TLC, HPLC and HPLC-MSn to characterize and identify the flavonoids. As a result, we established the one-dimensional TLC fingerprint, the two-dimensional TLC fingerprint and the HPLC fingerprint of the flavonoids. Identification and analysis of the flavonoids was carried out upon HPLC-MSn, and we successfully identified 11 main constituents according to the experiences that obtained in researchs of snow chrysanthemum and datas in relevant literatures, finally. In summary, this study laid a foundation for future research of snow chrysanthemum.
