Objective: Hippo signaling pathway is the pathway to control the organ size that found in Drosophila, Yap protein is the core component of this signal pathway. Hippo has relationsip with the Transformation between mammalian tissues and the external mechanicaltension. Wound healing and Hypertrophic scar involved with a series of complex cell biological processes.The formation of hypertrophic scars is associated with several factors, such as gender,age, physical fitness, infection,and so on.The pathology of the hypertrophic scars is the fibroblast proliferation and extracellular matrix depo- sition, and yap protein is closely related with the fibroblast growth factor secretion, cell proliferation and the tension from the scar. In this experiment,we present the expression location and change of yap protein through the time from the trauma to the Hypertrophic scar formation on the rabbit ear model. Based on this date to investigate the role of Yap protein in wound healing and hyperplasia scar, and provide a theoretical basis for the molecular level for the scar formation, and provide ideas and new therapeutic targets for the treatment of scar.Methods: a total of 10 New Zealand white rabbits, male or female, average weight 2.5-3.0Kg. Each rabbit left ear set to group A, the right ear is set to the B group, A group, eachrabbit ear punch 3 0.6cm diameter circu- cartilage, completely destroys cartilage membrane; B group, each rabbit ear pu- nch 3 1cm diametercircular lesions to cartilage, completely destroyed the perichondrium, each wound interval >1cm. Thereafter, each rabbit rearing conditions are the same, from near the head wound on the 14 day; 30 days take the middle wound; 60 days for the rest of my wound. The specimens were placed in 10% neutral formalin fixed for 12 hours, the conventional gradient ethanoldehydration, paraffin embedding slice, immunohistochemistry and histological staining.Immunohistochemical staining of Yap positive signals were brownish yellow granules. Yap image analysis method: 1, 400 times in each section were observed under the microscope inthe cells of epidermis and in the number of Yap positive granules, and calculate the. Eachslice observation 5 vision takes its mean comparison. 2, with the Image-pro6.0 image analysissoftware system, the expression intensity of Yap with a value of IOD indicates, each sectiontakes 5 view, each view were measured IOD values and take the mean number of samples asthe IOD value, comparison.Results: the success rate of a scar, model: A group’s success 24 scar, B group made Model27 scar(model success criteria: visible to the naked eye obvious wound tissue hyperplasia,qualitative hard, scar surface than the surrounding normal tissue is higher than-2mm);Two, under the microscope and Yap immunohistochemical staining results: 1, 14 days, Agroup and B group tissue around the wound to the wound center retraction, part of wound surface drying, granulation tissue, inflammatory cells and capillary angiogenesis under the microscope, the epidermal cells of Yap was mainly expressed in the cytoplasm, cytoplasmicvisible fine brown granules the number of fibroblasts, little, brown granules found in the nucleus of a few;Naked eye observation, 30 days, 2, wound healing and scar hyperplasia, hyperplasia scar the color red, hard, and B group than A group obviously wound scar hyperplasia, fiber mirror, scarsurface is a thin epidermis cells and associated histologically normal skin appendages such ashair follicles, sebaceous glands, such as the lack of scar, lower into fiber cell generation,spindle shaped and arranged in neat rows, brown yellow granules appeared in the fibroblast nuclei, the rest of the cytoplasm brown yellow stained naked eye observation; 3, 60 days, A group, part of scar is 30 days reduced the rest 30 days, lower vertical diameter increases, and the hardness increase, seen under the microscope, scar skin a lot of fibroblast cell generation,B group part of scar fibroblasts arranged in disorder, whorls form, individual scarvisible hyalinization. 4, 70-90 days of observation, the naked eye see scar narrow,microscope, the fibroblasts decreased, partially hyalinized. Three, analysis of data statistics: Statistical analysis of data: 1, 14 days, group B on the positive particles, integral optical density, mean optical density values are higher than those in group A, and P <0.05, the difference was statistically significant; 2,30 days, group B higher than the number of particles in the positive A group, P> 0.05, the difference was not statistically significant, integral optical density, the average optical density values are higher than those in group A, and P <0.05, the difference was statistically significant; 3,60 day, group B is higher than the number of particles in the positive A group, P> 0.05, the difference was not statistically significant, integral optical density, the average optical density values are higher than those in group A, and P <0.05, the difference was statistically significant; 4, the number of positive particles A group 14 days: 22.38 ± 5.12, the first 30 days: 34.88 ± 3.56, the first 60 days: 32.38 ± 5.82,30 days and 60 days values are higher than 14 days, P <0.05, the difference statistically significant, 60 days less than 30 days the value, P> 0.05, the difference was not statistically significant; Group B 14 days: 35.13 ± 6.12, the first 30 days: 39.63 ± 5.68, the first 60 days; 34.50 ± 2.4430 day value high in 14 days and 60 days, p> 0.05, the difference was not statistically significant, the value is less than 30 days 60 days, p> 0.05, the difference was not statistically significant; 5, the average optical density A group 14 days: 0.20 ± 0.01 The first 30 days: 0.24 ± 0.03, the first 60 days: 0.22 ± 0.06,30 day value greater than 14 days, p <0.05, the difference was statistically significant, 30 days value is greater than 60 days, P> 0.05, the difference was not statistically meaning; Group B Day 14: 0.22 ± 0.01, the first 30 days: 0.30 ± 0.04, the first 60 days: 0.31 ± 0.02,30 days more than 14 days, P <0.05, the difference was statistically significant, 30 days less than 60 days value, p> 0.05, the difference was not statistically significant; 6, the first 14 days IOD A Group: 1016.63 ± 90.07, the first 30 days: 1368.00 ± 169.01, the first 60 days: 1355.13 ± 162.87,30 days and 60 days values are more than 14 days, and p <0.05, the difference was statistically significant, 30 days value is greater than 60 days, P> 0.05, the difference was not statistically significant; B group Day 14: 1358.38 ± 186.57, the first 30 days: 1921.75 ± 100.76 The first 60 days: 1911.88 ± 114.00,30 days and 60 days value higher than 14 days, p <0.05, the difference was statistically significant, 60 days value is less than 30 days, P> 0.05, the difference was not statistically significant.
