Aconiti Lateralis Radix Praeparata (fuzi) is a famous medicinal material and commonly used as toxic traditional Chinese medicine. Fuzi is the processed product of the daughter root of Aconitum carmichaelii Debx. Fuzi native to Sichuan is yielded from Jiangyou、Anxian and Butuo in Sichuan, Hanzhong in Shanxi and Dali in Yunnan. Fuzi is pungent, sweet, hot, toxic and the channel-tropism of heart, spleen and kidney in properties and firstly recorded in 《ShenNongBenCaoJing》and highly praised for the first and top medicine in restoring Yang to save from collapse. The efficacies of fuzi are restoring Yang to save from collapse, tonifying fire to help Yang and dissipating cold to relieving pain and the fictions are debilitation Yang, cold limbs, micro pulse, impotence, cold palace, cold and pain heart, deficiency cold, edema Yin, deficiency Yang, cold and wet to pain and so on. Modern pharmacological studies have shown that fuzi have some functions of strong heart, anti-inflammatory, anti-tumor and anti-aging, sedation, analgesia, enhance immunity and clinical used in the treatment of acute myocardial infarction caused by shock, low blood pressure, coronary heart disease and angina pectoris.Fuzi contains a lot of compounds that complicated and it is important to establish the method of determination and comprehensively and scientifically recognize composition for the quality control and clinical application. The traditional processing method is complex in process, poor in controllability and leads to be greatly lost in effective components and severely remained in " danba", so there is a great controversy in the quality, effectiveness and safety of fuzi.According to the above problem, this paper conducted the following study:(1) The study on determination offuziThe study on the method of determination of water-soluble components in fuzi:This is first report of quantitative analysis for fuzi by the simultaneous determination of five water-soluble components.Optimization of the method of determination on ester-type alkaloids in fuzi:When the samples were extracteded with HCI solution (0.05mol·L-1) by sonication, the contents of six ester-type alkaloids were about15~40%higher than which extracted with method of ChP. Vol I (2010Edition).The study on stability of the solutions extracted from fuzi:To study the dynamic variation patterns and stabilities of ester aconitum alkaloids in solutions extracted from fuzi during heating process, the results showed that the ester alkaloids were stable in acid conditions and unstable in alkaline conditonns.(2)The study on the modern processed methodsThe processing method recorded in Chinese Pharmacopoeia (2010) is complex in process, poor in controllability and leads to be greatly lost in effective components and severely remained in "danba",so there is a great controversy in the quality) effectiveness and safety of fuzi. To explore new processing methods of fuzi, this study proposed the processing methods and equipments of dry-heat sterilization and wet-heat sterilization used in pharmaceutical industry. The new processing method need not be used the water of "danba" to moisten, soak, boil and rinse to avoid the losing of effective components and is simple, efficient and controllable.(3)The study on HPLC-MS in fuziThe analysis of the compounds in fuzi by HPLC-Q-TOF-MS:The study identified totally36compounds by HPLC-Q-TOF-MS to comprehensively analysis the chemical composition.The analysis of the variation of chemical composition in the process of decoction in fuzi by HPLC-MS:The study explored the law of the variation of25compounds in the process of decction, the results showed that double-type ester alkaloids reduced all the on and few in2h; single-type ester alkaloids firstly increased, then gradually deceased and reached the peak in6to8h; alcohol amine-type alkaloids increased all the on; general-type alkaloids firstly increased, then slightly reduced and almost balance and reached the peak in6to8h.
