| Background: Atherosclerosis (AS) poses a most serious threat to human’shealth at present.According to WTO investigation,AS caused Cardiovascular andCerebrovascular diseases has become a trend that the morbidity and mortalityof these diseases are on the rise.Atherosclerosis can be triggered by a widevariety of factors.With the booming of the socioeconomy,it is largelyattributable to changes in Chinese diet,the faster pace of life and mental andsocial states,coupled with other factors such as genes, environment andmetabolism and so on.Some researches show that dyslipidamia is the most typicalrisk of all. So impactfully controlling dyslipidamia is very important for theprevention and treatment of cardiovascular disease.The understanding ofatherosclerosis in Traditional Chinese medicine (TCM) records some similarsymptoms which described in chest impediment, true heart pain, turbidphlegm,dizziness.Especially, chest impediment and true heart pain registercharacteristics similar to those of the cardiovascular disease.The diseaseinvolves complex etiologies and pathogenesis,TCM attributes the diseaseprimarily to improper diet, emotional disorder, fatigue and internal damage andweakness generally. According to TCM,in most cases harms zangfu organs such asheart, liver, spleen,lung and kidney.And in the theory of TCM,the disease iscaused by deficiency in origin and excess in superficiality, deficiency-excessmixing,Deficiency in origin reflects the deficiency in any of these five organs.Excess in superficiality describes pathologic factors such as turbid phlegm,stagnation of Qi,cold coagulation and blood stasis.Purpose:Using acute toxicity test in mice,to confirm the maximum dosage ofQingzhitongmai Granules.Observing the anti-hyperlipoidaemia anti-atherosclero-sis effects of Qingzhitongmai Granules and further study its correspondingmechanisms by means of modelling the hyperlipidemia of mice and rabbits,and therefore it will provide secure and theoretical guidance for the clinicalapplication of Qingzhitongmai Granules.Methods:1.To make sure the the maximum dosage of Qingzhitongmai Granules by means ofConventional maximum dosage.2.Using vitelline normal saline to build hyperlipidaemia model of mice;Hyperlipidaemia rabbits model was established using high-fat diet.3.Compared with the positive control drug named Xuezhikang,observing the effectsof Qingzhitongmai Granules on every indicator of dyslipidemia in experimentalanimals.4.Observing the effects of Qingzhitongmai Granules on aorta atheroscleroticplaques of domestic rabbits.2.As the establishment of the model of hyperlipemia mice successfully,Qingzhiton-gmai GranulesResults:1.Mice in acute toxicity test have no abnormal activity and weight etc,and thereis no mortality in test cycle.2.As the establishment of the model of hyperlipemia mice successfully,Qingzhiton-gmai Granules can significantly decreased the elevated level of total chole-sterol(TC) and triglyceride (TG),and have similar pesticide effect with thepositive control drug Xuezhikang and Simvastatin.3.Qingzhitongmai Granules can significantly decreased the elevated level oftriglyceride (TG),and have similar pesticide effect with the positive controldrug Xuezhikang,according to successfully building the high-fat diet in domesticrabbits.The effects of Qingzhitongmai Granules on total cholesterol(TC), lowdensity lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholestero(HDL-C) were not obvious.4.Qingzhitongmai Granules delays or regress the progression of aortaatherosclerotic plaque of rabbits. Conclusion:1.The maximum dosage of Qingzhitongmai Granules was40g/kg/d,equivalent to crudemedicinal172.8g/kg/d.2.Qingzhitongmai Granules can significantly decreased the level of triglyceride(TG) in the experimental animals.3.Qingzhitongmai Granules delays or regress the progression of aorta atheroscle--rotic plaques of rabbits,then get the effect of Anti-atherosclerosis.4.Mechanism fuction of Qingzhitongmai Granules may be by lowering blood lipid,so that reducing damage of structure and function of endothelial cells liningblood vessels because of abnormal lipid, then restoring the blood vessel functionand stabilizing atherosclerotic plaques,finally restraining or alleviating theoccurrence and development of cardiovascular disease. |
