Purpose:To experiment in isoproterenol induced myocardial ischemia in ratsas a model, to observe the acupoint on myocardial tissue of rats with chloride(CL-) channel effect of PKC, PKG protein expression and CFTR, CLCa1geneexpression and protein kinase, to investigate the effect of specific acupointsalong meridians mechanism, to provide scientific experimental basis foracupuncture treatment of myocardial disease.Material and method:70SD rats,randomly take10for the blank group,and randomly divided the others into4groups:blank,modeling,Neiguan,Lieque,and Non-acupoint,and make them myocardial ischemia models by the treatment withmultiple subcutaneous injection of isoproterenol (ISO),except the blank group.Provide the electro-acupuncture treatments on Neiguan,Lieque,Non-acupoints(themiddle of Tianshu and Shenque points)separately to these three groups,oncea day for7days.Then observe cardiac tissue were observed by HE staining,with BL-420S biological experimental system to detect the ECG of rats in eachgroup,as compared to mV number counting index.Using RT-PCR(Realtime-PCR)indetection of myocardial tissue in rats of chloride channels in cystic fibrosistransmembrane conductance regulator(CFTR),the expression level of CLCa1geneexpression;Western level by Blot blotting in myocardial tissue of rats withchloride channel protein kinase PKC,protein PKG.Results:1.Compared with the normal group, model group, Lieque group and non ECG and nonacupoint group myocardial cells have changed, no statistically significantdifferences in measurement data but (P>0.05);compared with the model group,Neiguan group had obvious improvement, statistically significant differences in measurement data (P<0.05).2.Compared with the normal group, model group, Lieque acupoint and non acupointgroup PKC protein kinase PKG, myocardial protein rats increased significantly,the difference was statistically significant (P<0.01);compared with the modelgroup, Neiguan PKC protein kinase PKG, protein expression of rat myocardial grouprise high, the difference was statistically significant (P<0.05).3.Compared with the normal group, model group, listed lack of acupuncture pointgroup, non chloride channel in myocardial tissue of rats with non acupoint groupin CFTR,CLCa1gene expression increased,the difference was statisticallysignificant (P<0.01);compared with the model group, the chloride channel inmyocardial tissue of rats in CFTR, CLCa1gene at the expression decreased, thedifference was statistically significant (P<0.01).Conclusion:1.It effectively suppress the pathological changes of heart tissue withacupuncture at Neiguan acupoint of myocardial ischemia rats.2.Acupuncture Neiguan point can be elevated myocardial protein kinase PKG,PKCprotein expression,has a protective effect on myocardial injury.3.Acupuncture Neiguan point can regulate the content of chlorine ion channelprotein expression of CLCa1,CFTR,thereby regulating myocardial tissue chloridechannels,which may be one of the mechanisms of myocardial injury caused bymyocardial ischemia.4.The effect of Neiguan point tired specificity is exist....
