Purpose:The study tries to observe the effect on acute cholecystitis of guineaby triple needling the lower confluent point of biliary, and explores itsmechanism through observe the change of β-endorphin(β-EP) and cholecystokinin-octopeptide (CCK-8)’s content as well as the peripheral blood WBC number (WBC),providing a scientific and reasonable experimental basis for the treatment ofacute cholecystitis with a better needling method in clinic.Material and method:Totally80healthy male guinea pigs of clean grade wereselected, weighting350-400g. Placed them in quiet illumination at7:00-19:00,and domesticated them in the dark laboratory at19:00-7:00for one week,andperformed the formal experiment after acclimatization.The guinea pigs weredivided into5groups: normal group with10guinea pigs、model group with10guinea pigs、drug group with20guinea pigs、acupuncture group with20guineapigs and triple needling group with20guinea pigs.5guinea pigs were fed inone cage both in normal group and in model group, and1guinea pig was fed inone cage in other groups. After be improved, applied Duchenne acute cholecystitismodel method to make models,and the therapeutic means were,in normal group: fednormally under the same condition without any processing; in model group: fednormally after making models successfully;in drug group: gave0.007mlintraperitoneal injection (administration after distilled water was diluted)with compound diclofenac sodium and lidocaine hydrochloride injection(2ml/injection,including75mg diclofenac sodium、20mg lidocaine)in the caseof guinea pigs awake after making models successfully; in acupuncture group:after making models successfully, selected bilateral GB34in the case of guineapigs awake, in accordance with “Map for acupoints of experimental animals”enacted by the national acupuncture and moxibustion institute-experimental acupuncture research association,and penetrated into3mm-5mm with0.25mm×15mmfine needle, reforcing and reducing after developing needle sensation, retainingneedles for30minutes, manipulating needles5times during treatment period.Then withdraw the needles, once a day, and continued treating for5times; intriple needling group: penetrated GB34into3mm-5mm with0.25mm×15mm fineneedle, after that, acupunctured the upper (proximal end) and lower (distal end)point along the gallbladder meridian with fine needles. Peripheral blood WBCnumber (WBC): test the peripheral blood WBC number at pre-experiment, aftermaking models and post-treatment. At the end of the treatment, sacrificed theguinea pigs in each group, and fresh blood、homogenate were taken for theexamination of the β-EP and CCK-8content with radiation therapy.Results:1.Change of β-EP and CCK-8’s content in guinea pigs’plasmaContrast between model group and normal group, the content of β-EP in plasmasignificantly increases (p<0.01).The content of β-EP in drug group、acupuncturegroup and triple needling group are significantly higher than that in model group,which has significant difference (p<0.01).There is no difference between druggroup and acupuncture group (p>0.05);contrast with acupuncture group,the tripleneedling group has significant difference (p<0.01).Contrast between model group and normal group, the content of CCK-8in plasmaare obviously increased (p<0.01). The content of CCK-8in drug group、acupuncturegroup and triple needling group are obviously lower than that in model group,which has significant difference (p<0.01); there is no difference between druggroup and triple needling group (p>0.05);contrast with acupuncture group,thetriple needling group has statistical significance (p<0.05).2. Peripheral blood WBC numberPeripheral blood WBC number before the experiment: there is no differenceamong these groups (p>0.05).Peripheral blood WBC number after making models: contrast with normal group, the number of WBC in other groups are obviously increased, which has significantdifference (p<0.01).Peripheral blood WBC number after treatment: contrast with model group, thenormal group has significant difference (p<0.01); Peripheral blood WBC numberin drug group、acupuncture group and triple needling group are obviously lowerthan that in model group, which has statistical significance (p<0.01); contrastwith drug group and acupuncture group, the triple needling group has significantdifference (p<0.01);contrast with acupuncture group,the triple needling grouphas statistical significance (p<0.05)..Conclusion:1. The drug group、acupuncture group and triple needling group all have an effecton treatment for models of guinea acute cholecystitis.2. Triple needling GB34can increase the content of β-EP in plasma and decreasethe content of CCK-8, alleviate the response for inflammation. Suggest thatacupuncture has the function of anti-inflammatory, immune and analgesia relyingon the attendance of opioid peptide system and brain-gut peptide; it can alsoshow that acupuncture not only has the effect of anti-inflammatory and immunebut also can alleviate the pain caused by inflammatory, having the effect ofanalgesia.3. Triple needling GB34has a better effect than that with normal acupuncture.
