Purpose: This research through the rat model of chronic heart failure after filling Yiqi huoxue Compound and lisinopril tablet, explore the effect on chronic heart failure rat of Yiqi huoxue Compound of myocardial energy metabolism, make the PGC-1α、NRF-1、mt TFA、ATP as the basis, discuss the related mechanism and targets, for the motherland medicine will provide a scientific basis for prevention and treatment of chronic heart failure failure and experimental method.Material and method: 1 To established the Chronic heart failure rat model:Choose SPF 12 weeks healthy SD rats, male, 80, in 250±10 g weight,by ligation of left anterior descending coronary artery,cooperate with swimming exhaustion type and the method of dieting to established the Chronic heart failure rat model. In the treatment group compared with model group, treatment group BNP concentration decreased, p = 0.000(p < 0.01) and significant difference between groups, explains the experimental drug therapy to reduce the BNP. Compared with model group, treatment group of NTF- 1 、mt TFA m RNA expression and protein expression are higher,(p = 0.000 < 0.01) and significant difference between groups, suggests that the treatment group of drugs can improve the myocardial tissue of NTF- 1, the content of mt TFA prompts the experimental drug treatment can increase the generation of mitochondrial。 4 Observation under electron microscopy(sem) in the myocardial mitochondria that: normal group in the rat myocardial mitochondria in cells abundance, shape size, ridge structures such as can be clearly seen in the mitochondria, mitochondrial outer membrane smooth and no damage, no swelling of mitochondria; Model group rats myocardial mitochondria in cells is relatively fewer in number form size is irregular, mitochondria swelling, ridge in the mitochondria structure such as texture disorder is not clear, even form only frame structure of vacuoles, mitochondria mitochondrial outer membrane damage, even fracture; Western medicine group in the rat myocardial mitochondria in cells compared with model group increased number, but it is still less than normal group, although mitochondrial abnormalities, such as size of irregular form, but the overall shape size, mitochondrial cavitation abnormal mitochondria structure to reduce in the model group; Chinese traditional medicine group in the rat myocardial mitochondria in cells number less than western medicine group, but more than the model group, the mitochondrial cavitation, the swelling deformation, spinal disorders, such as abnormal structure compared with model group decreased. Instructions, yiqi huoxue compound can optimize the proliferation of mitochondria, improve the abnormal mitochondria structure, group and western medicine and Chinese medicine treatment group.Conclusion: 1 By comparing the data, found that the content of ATP and the NRF- 1, mt TFA reduced expression in the process of chronic heart failure, heart failure happened when there exists a mitochondrial energy metabolism disorder. 2 Through the comparison between each group, the microscopic microstructure change of mitochondria, prove that in the presence of abnormal mitochondria damage in heart failure, compared with model group, the experimental drug on mitochondria have good curative effect. 3 By comparing the results between groups that yiqi huoxue compound is able to improve myocardial energy metabolism, the formulas by increasing NRF- 1、 the expression of mt TFA,increase grain with generation and ATP production capacity, improve myocardial energy metabolism, inhibit the action of the ventricular remodeling.
