Purpose: This research inquires into the effect of acupuncture combined with ginseng decoction on chronic fatigue rats saliva s Ig A contens, NK cell activity and function of phagocyte,revealing acupuncture combined with medicine on s Ig A and nonspecific immune intervention mechanism in the state of fatigue, thus provides the theory basis for simultaneously improving immunity and anti-fatigue.Material and method:65 SPF male Wistar rats were randomly assigned into 5 groups(n=13 each): blank control group, model group, medicine group,acupuncture group, acupuncture combined with medicine group. The blank control group was not treated, while the remaining four groups were adopted exhaustive swimming plus restraint method for two weeks. After copying the model, the blank control group and the model group rats were given hand grip every day; the acupuncture group was applied subjects with electro-acupuncture on the bilateral acupuncture points "Xinshu"(BL15), "Pishu"(BL20) and "Shenshu"(BL23) for 20 minutes once a day for a week; medicine group was given ginseng decoction gavage 1.5m L once a day for a week; acupuncture combined with medicine group was simultaneously applied acupuncture and ginseng decoction gavage two factors once a day for a week. After the end of treatment, indicators were detected in rats. Each group had 8 rats by intraperitoneal injection of 20% chicken red blood cell suspension 1m L, anesthetized after static 30 minutes, intraperitoneally injected Sodium Chloride Physiological Solution 2 m L, gently massaged abdominal for 1 minute and cut abdomen, suck out the peritoneal fluid 1m L, treated under the microscope and selected 100 phagocytic cells, numbered phagocytic cells which contained chicken red blood cells and calculated phagocytic percentage; at the same time, we picked out each rat’s salivary glands, using ELISA to detect the content of s Ig A in saliva. The remaining 5rats in each group were applied aseptic spleen lymphocyte extraction operation, we used MTT method to detect the activity of NK cells.Results:1 Rat general situation: The color of hair, the diet and the behavior of rats in blank control group were normal. After modeling, rats appeared apathetic, slow activity, loose stool, emaciated, fur is not glory, easy get together and irritable, etc. After acupuncture, ginseng decoction, acupuncture combined with medicine treatment, the symptoms of rats in each group were improved. 2.Effect on the content of salivary s Ig A: Compared with the blank control group, the content of rats salivary s Ig A in model group decreased(P<0.01);compared with the model group,the content of s Ig A in all treatment groups were higher than that of model group(P<0.05); in comparison among the treatment groups, no significant difference between medicine group and acupuncture group(P>0.05), nevertheless acupuncture combined with medicine group made a significant difference(P<0.01) compared with acupuncture group and medicine group and had the best effect. 3.Effect on phagocytic function: Compared with the blank control group, phagocytic percentage of model group decreased(P<0.01); compared with the model group, percentage of phagocytic cells in treatment groups were higher than that in model group(P<0.05); in comparison among the treatment groups, no significant difference between medicine group and acupuncture group(P>0.05), nevertheless acupuncture combined with medicine group made a significant difference(P<0.01) compared with acupuncture group and medicine group and had the best effect. 4.Effect on NK cell activity: Compared with the blank control group, the activity of NK cells in model group decreased(P<0.01); compared with the model group, the activity of NK cells in treatment groups were higher than that of model group(P<0.05); in comparison among the treatment groups, no significant difference between medicine group and acupuncture group(P>0.05), nevertheless acupuncturecombined with medicine group made a significant difference(P<0.05) compared with acupuncture group and medicine group and had the best effect.Conclusion:1.The effect of acupuncture combined with medicine on fatigue resistance is remarkable. 2.The change of immune function may be one of the mechanisms of chronic fatigue. 3.Acupuncture combined with medicine can improve content of saliva s Ig A. 4.Acupuncture combined with medicine can improve the non-specific immune function. 5.Acupuncture combined with medicine can effectively antagonize coupling reaction of fatigue induced immune function decreased.
