Objective: Based on the potential trend of acupuncture and moxibustion treatment on anti-fatigue, this experiment which acupunctured Xinshu, Pishu and Shenshu combined with Ginseng decoction expected to carry out experimental study on the rats’ humoral immune effects in chronic fatigue model group. Explore the principle of acupuncture combined with medicine treatment on anti-fatigue for acupuncture combined with medicine applied clinically.Expect it could play an important role in the treatment on anti-fatigue.Materials and methods:Five groups of experimental animals. 40 Wistar male rats of 7 weeks old were randomly divided into five groups: control group, model group, Chinese medicine group, acupuncture group, acupuncture combined with medicine group, 8 rats in each group. A chronic fatigue model was prepared in a current widely accepted way called exhaustive swimming model plus the fatigue change induced by bound cylinder. After the establishment of the model, the acupuncture group adopted acupuncturing acupoints of Xinshu, Pishu and Shenshu for 20 min every time, lasted for 7 days; Chinese medicine group daily 1.5ml of 100% Ginseng decoction gavage for 7 days; acupuncture combined with medicine group, by acupuncturing Xinshu, Pishu and Shenshu plus 1.5ml of 100% ginseng decoction gavage once a day for 7 days. After the end of the experiment, with 10% chloral hydrate anesthetized rats,then suck out the blood from the abdominal aorta and cryopreserved. ELISA was used to detect serum Ig A, Ig G, Ig M. While HC50 method for the content of serum hemolysin.Results: 1. Rat Ig A content determination After 7 days of treatment, the content of Ig A in the serum of rats in each group were detected, the detection results showed that compared with the blank group, serum Ig A content from model group, Chinese medicine group, acupuncturegroup, acupuncture combined with medicine group were significantly decreased(P < 0.01). Compared with the model control group, the content of Ig A in serum of rats in Chinese medicine group, acupuncture group and acupuncture combined with medicine group were significantly higher than that in model group(P<0.01). The comparison between the treatment groups showed that the acupuncture combined with ginseng decoction group was significantly higher than the acupuncture group and the Chinese medicine group(P<0.01). 2. Rat Ig G content determination Compared with the blank group, the content of Ig G in the serum of the model group, the Chinese medicine group, the acupuncture group and the acupuncture combined with ginseng decoction group were significantly decreased(P<0.01). Compared with the model control group, the content of Ig G in serum of Chinese medicine group, acupuncture group and acupuncture combined with Ginseng decoction group were significantly higher than that in model group(P<0.01). The content from the acupuncture medicine group was significantly higher than that in the Chinese medicine group and the acupuncture group(P<0.01). There was no significant difference between the Chinese medicine group and the acupuncture group(P>0.05). 3. Rat Ig M content determination Compared with the blank group: model group, Chinese medicine group, acupuncture group, serum Ig M levels were lower than that of the control group(P < 0.05), Ig M content in serum of rats in the acupuncture group was close to the blank group and had no statistical difference(P > 0.05). Compared with the model group, there was no significant difference between the Chinese medicine group and acupuncture group(P>0.05), and the content of Ig M in the serum of the acupuncture combined with medicine group was significantly higher(P<0.01). 4. Determination of serum hemolysin content in rats Compared with the blank group: model group, Chinese medicine group,acupuncture group, acupuncture and medicine group rats serum hemolysin were significantly lower than that in blank control group(P < 0.01). Compared with the model controlroup: Chinese medicine group, acupuncture group, acupuncture combined with medicine group rats serum hemolysin contents were significantly higher than that in the model group(P < 0.01). The comparison between the treatment groups showed that the acupuncture group and the acupuncture combined with medicine group were higher than that of the Chinese medicine group(P<0.05). There was no significant difference between acupuncture group and acupuncture combined with medicine group(P>0.05).Conclusions: 1.Chinese medicine, acupuncture, acupuncture combined with Ginseng decoction could improve the chronic fatigue rat Ig A content and the acupuncture alone effected better than Chinese medicine alone. While acupuncture combined with Ginseng decoction worked best, but none of them reached the normal level. 2.Chinese medicine, acupuncture, acupuncture combined with Ginseng decoction could increase the chronic fatigue rat Ig G content, acupuncture combined with Ginseng decoction worked best, but none of them reached the normal level. 3.Neither Chinese medicine nor acupuncture alone could increase chronic fatigue rat Ig M content,however acupuncture combined with Ginseng decoction could obviously improve the chronic fatigue rat Ig M content, and the effect basically reached the normal level. 4.Chinese medicine, acupuncture, acupuncture combined with Ginseng decoction all could increase serum hemolysin content in rats.Acupuncture, acupuncture combined with Ginseng decoction worked better than Chinese medicine alone, but none of the them reached the normal level. 5.Acupuncture combined with Ginseng decoction worked better on humoral immune regulation induced by chronic fatigue than acupuncture alone or Chinese medicine alone.The reason might be that acupuncture could combine with Chinese medicine and work together smoothly in improving the humoral immune regulation.
