| Objective: Observed the beneficial effect of acupuncture combined ginseng therapy against exercise-induced fatigue from the perspective of skeletal muscle-central nerve-energy supply,and elucidated its effect mechanism from the perspective of flora-metabolism,to provide scientific support for acupuncture combined ginseng against exercise-induced fatigue.Methods: I.Clinical studySixty healthy males who matched the inclusion criteria were separated into control,electroacupuncture,ginseng,and acupuncture combined ginseng groups.The four groups were treated five times each week for six intervention sessions.Assessment variables were exercise completion time,average heart rate,heart rate variability,flash fusion frequency,upper limb grip strength,scale for Rating of Percerived Exertion,stroop test,blood lactate,and hemoglobin.In terms of flash fusion frequency,grip strength,subjective fatigue sensation scale,stroop test,and blood lactic acid,the acupuncture combined ginseng has the greatest effect compared to other single therapies,which is primarily reflected in the immediate recovery after exercise.II.Experimental studyEighty male C57/BL6 SPF mice were separated into four groups: control,electroacupuncture,ginseng,and acupuncture combined ginseng.Mice in each group were treated to a fatiguing stick-turning activity every 10 days,and their grip strength was measured 30 days after completion.The four groups were administered medication once every day for 30 days,and the mice were collected feces and serum after 20 and 30 days.The expression of myosin protein and laminin-2 protein was detected by immunofluorescence double-labeling,the key proteins of the AMPK/PGC-1 signaling pathway were analyzed by Western Blot,the expression of central excitatory neurotransmitters DA and EPI and the body energy metabolism supply substances AMP,ATP,PK and SDH were detected by ELISA,and the intestinal flora and thanksomics were also employed to screen the differential flora Intestinal flora and metabolites were also utilized to search for changes in flora and metabolites and examine probable reasons.Results: I.Clinical studiesIn terms of completion time at different exercise distances,the electro-acupuncture group had no significant modifying effect,whereas the ginseng and acupuncture combined ginseng groups had significant enhancing effects,with the acupuncture combined ginseng superior to ginseng.In terms of mean heart rate,heart rate variability,and haemoglobin content,there were no significant differences between the groups;however,in terms of flash fusion frequency and grip strength,there were statistically significant differences between the groups.In terms of flash fusion frequency,grip strength,subjective fatigue perception scale,executive ability test,and blood lactate.however,the combination of needle and medication had the best impact compared to other monotherapies,which was mostly apparent in the fast recovery following exercise.In terms of completion time under different exercise distances,the electroacupuncture group had no significant regulatory effect,while the ginseng group and the acupuncture combined ginseng group had significant improvement effects,the acupuncture combined ginseng e being superior to ginseng.There were no significant differences among the groups in the mean heart rate,heart rate variability,hemoglobin content and other indicators.However,there were significant differences between the electroacupuncture group and the ginseng group in terms of completion time under different exercise distancesII.Experimental investigations1.Grasp strength,Skeletal Muscle Skeletal Protein,and AMPK/PGC-1 Prior to and Following Exercise After treatment with electroacupuncture,ginseng,or acupuncture combined ginseng,each group of mice showed considerable improvement in terms of energy metabolism signal pathway proteins,with the improvement in the acupuncture combined ginseng group being the most pronounced.Regarding central neurotransmitters and substances involved in the body’s energy metabolism,the regulatory advantages of each category vary depending on the intervention time.Electroacupuncture treatment is reflected in the excitatory central neurotransmitters.The energy metabolism of the body reflects the ginseng group,but the overall regulation of the acupuncture combined ginseng group is more evident.In terms of central neurotransmitters,the levels of EPI and DA in the acupuncture and drug combination group were higher than in the control group,the electroacupuncture group,and the ginseng group;In terms of energy metabolism substances,the levels of AMP,ATP,PK,and SDH in the acupuncture combined ginseng group were also higher than in the other groups.These results suggest that the acupuncture combined ginseng and medication can increase the exercise capacity of mice with exercise-induced tiredness via a variety of physiological pathways.2.Each therapy can better regulate the structure of the intestinal flora and correct the flora imbalance in mice with exercise-induced tiredness.Streptococcus and Fasciococcus upregulation in three groups and identification of Unidentified_.Under the trend of downregulation of [Paraprotetellaceae] bacteria and Lactococcus,the regulatory effect of acupuncture combined ginseng therapy is superior to that of other single therapies,with Salmonella,Streptococcus,and Butyrococcus being the predominant bacteria.3.In combination therapy with acupuncture and medication,in addition to lipids and lipid molecular metabolites,the most regulated metabolic products have unique regulatory effects on nucleosides,nucleotides,and analogues.Its distinctive metabolic pathways include arginine biosynthesis and pyrimidine metabolism.Conclusion: 1.The acupuncture combined ginseng can significantly improve the physical function of the exerciser,shorten the exercise completion time,and have better immediate recovery effects in terms of muscle strength,psychological fatigue,central nervous and mental fatigue,and lactic acid metabolism accumulation than other single intervention therapies,but there is no advantage in improving heart rate,heart rate variability,and aerobic metabolism.2.Acupuncture combined ginseng therapy has the beneficial regulating effect of acupuncture on excitatory central neurotransmitters such as DA and EPI,as well as ginseng on energy metabolism supply chemicals such as AMP,ATP,SDH,and PK,and can activate AMPK/PGC-1 to a higher extent.Signal pathways that promote the production of myosin protein in skeletal muscle and improve muscle strength have an onset time that is faster than that of other monotherapies.3.By controlling Sardinia,Streptococcus,and Butyrococcus,acupuncture combined ginseng therapy may correct the gut flora imbalance in mice with exercise-induced weariness.Pathways of arginine biosynthesis and pyrimidine metabolism may contribute to the enhanced efficacy of acupuncture combined ginseng. |
