| Purpose:This experiment aims at demonstrating the comparison of different effects curing sciatic nerve damage between neural stem and muscle( non-neural stem) via Electronic-acupuncture. There is no doubt that using Electronic-acupuncture to cure peripheral nerve has remarkable effect. In this experiment Electronic-acupuncture can be used to acupuncture different tissue corresponding Huantiao-neural stem and muscle, and then the different effects curing sciatic nerve damage will be emerged through two ways.One is via observing the effect of sciatic nerve injury and repair.The other is through observing the different expression between the growth-protein GAP-43 associated sciatic nerve injury and tubulin MAP-2’s.Material and method:84 SD healthy rats in SPF level, whose weight and ages are around approximately 200±20g and 2-3 months, both number of male and female are 42. Base on randomly table, rats can be divided into seven groups, blank group, model clamped group, transected model group, jaw deep puncture(neural stem) group, clamp shallow stab(muscle), transverse deep puncture(neural stem) group, shallow stab transected(muscle) group, and there are 12 rats in each group. Using clamped and transected ways to make model in condition of sciatic nerve injury. Blank group is fed normally without any treatment. After successful modeling, the model group maintain in the same condition without any therapy treatment. Three days after a successful modeling, deep and shallow needling acupuncture group use Electronic-acupuncture to acupuncture Huantiao Neural stem and muscle tissue under Huantiao. Treatment lasts ten days, each time is 20 minutes.After ten days of treatment, exposing sciatic nerve in broken side, and then cut 2mm sciatic nerve from top and under respectively including crush, additionally, picking gastrocnemius of rats’ as well.Behavior by the swash plate assay to study the recovery of motor function in rats after Electronic-acupuncture at different tissue of Huantiao; Using Electronic scales to measure bilateral gastrocnemius wet weight, study in rats Electronic-acupuncture atrophy and recovery of muscle tissue after intervention byElectronic-acupuncture; Morphology with HE staining to observe morphological changes of damaged sciatic; By immunohistochemistry and western blot method to detect the expression of the damaged nerve segment between growth associated protein GAP-43 and microtubule-associated protein MAP-2, and then do statistical analyzing.Results:1.Experimental observation of the swash plateAs a consequent of experimental observation of the swash plate, compared with the blank group, the inclination angle of the two models experimental rats is lower swash plate and the difference is statistically significant(p<0.01). It implies that rats are liable to have motor dysfunction after prompted sciatic nerve damage; Compared with clamp model group, there is significant decline in terms of transected model group, the difference is statistically significant(p<0.01); After acupuncture treatment, compared with transected model group, plate tilt angle has been improved between transected shallow needling, crossing deep needling group of experiments in rats, and the difference is statistically significant(p<0.01); Compared with shallow transverse acupuncture group, improvement of transverse swash plate rat experiments and the inclination angle of the deep needling group are different, the difference is statistically significant(p<0.05); Compared with the model group clamp, Clamp shallow acupuncture group, deep puncture clamp experiments in rats inclination angle of the swash plate has improved significantly, the difference was statistically significant(p<0.05); Compared with shallow jaw acupuncture group compared, inclination angle of the swash plate acupuncture group and jaw deep upgrade rat experiments are different, the difference is statistically significant(p<0.05); Within both the transverse clamping deep needling group and deep needling group, which is close to the level of the blank group.2.Gastrocnemius wet weight recovery rateGastrocnemius wet weight results illustrate that: Compared with the control group, both models rats ipsilateral gastrocnemius wet weight have a certain decline, the difference is statistically significant(p<0.01); Compared with the model group clamp, lower transected model group more obvious difference was statistically significant(p<0.01); After acupuncture treatment, rats ipsilateral gastrocnemius wet weight and recovery rates have increased, compared with the cross-sectional model group, acupuncture group crossing shallow, deep needling group transected rats ipsilateralgastrocnemius wet weight and the recovery rate are significantly different, the difference is statistically significant(p<0.01); Acupuncture group compared with the transverse shallow, it differs in ipsilateral gastrocnemius wet weight recovery rate of deep needling group transected rats, the difference is statistically significant(p<0.05); Compared with the model group clamp, clamp shallow acupuncture group, clamping deep needling group are significantly different in ipsilateral gastrocnemius wet weight and the recovery rate, the difference is statistically significant(p<0.01); Compared with shallow jaw acupuncture group compared, difference exists in jaw deep needling group of rats’ ipsilateral gastrocnemius wet weight recovery, the difference is statistically significant(p<0.01).3.HE staining sciaticSciatic HE staining indicate:Sciatic nerve axons in the control group and Schwann cells arranged in neat rows; Both models group within axons and Schwann cells irregularly arranged, swelling, ulceration, and even the crack phenomenon have existed in Schwann cell, transected model group, even in the jaw model group; After acupuncture treatment, each group arranged axons and Schwann cells, swelling degeneration were improved, crossing deep needling group is better than the transverse superficial needling and deep needling group is better than the clamping jaw shallow needling.4.ImmunohistochemistryGrowth-associated protein GAP-43: blank group GAP-43 expression in the sciatic nerve a few positive nuclei blue, brown cytoplasm; Compared with the control group, the rest of the sciatic nerve in rats GAP-43 protein levels increase significantly different, the difference is statistically significant(p<0.01); After acupuncture treatment, compared with transected model group, crossing deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Acupuncture group compared with the transverse light, crossing deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Compared with the model group clamp, clamp shallow acupuncture group, clamping deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Shallow acupuncture group compared with the jaw, jaw deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01).Microtubule-associated protein MAP-2: the control group in MAP-2 expression in the sciatic nerve a few; compared with control group, the other groups sciatic nerve in MAP-2 protein levels are significantly different increases, the difference is statistically significant(p<0.01); After acupuncture treatment, compared with transected model group, crossing deep needling group regulated protein levels are significantly different, and the difference is statistically significant(p<0.01); Acupuncture group compared with the transverse light, crossing deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Compared with the model group clamp, clamp shallow acupuncture group, clamping deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Shallow acupuncture group compared with the jaw, jaw deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01).5.Western blot detectionGrowth-associated protein GAP-43: blank group GAP-43 expression in the sciatic nerve a few; Shallow acupuncture group compared with the jaw, jaw deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.05).Microtubule-associated protein MAP-2: the control group in MAP-2 expression in the sciatic nerve a few; compared with control group, the other groups sciatic nerve in MAP-2 protein levels are significantly different increases, the difference is statistically significant(p<0.01); After acupuncture treatment, compared with transected model group, crossing deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Acupuncture group compared with the transverse light, crossing deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Compared with the modelgroup clamp, clamp shallow acupuncture group, clamping deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01); Shallow acupuncture group compared with the jaw, jaw deep needling group regulated protein levels are significantly different, the difference is statistically significant(p<0.01).Conclusion:1. Acupuncture Huantiao different tissues, which can improve sciatic nerve injury in terms of rats gastrocnemius muscle, prevent muscle atrophy, and promoted recovery rates in rats with sciatic nerve injury in sports, consequently deep needling group is better than shallow acupuncture group.2. Acupuncture Huantiao different tissues, which can reduce the degeneration, death of neurons and promote axonal growth, additionally, in terms of accelerating the repair of damaged nerve,deep needling group is better than shallow acupuncture group.3. Acupuncture on neural stem,which can improve the damaged nerve micro-environment and promote the recovery of nerve damage,may be one of the mechanisms to repair peripheral nerve damage. |
