Objectives:The research is to study the effects that acupuncturing neural stem relatedacupoint--GB30tissues works on neuro-electrophysiology and c-fos after rat sciatic nerves(SNI) acute injury, and then compare the difference of effects of sciatic nerves acute injuryrepairing between acupuncturing GB-30neural stem and other tissues. To provideexperimental basis for scientific and reasonable choosing acupoint to acupuncture peripheralnerve injury; theory basis for studying neural stem group prescription for treating peripheralnerve injuryMaterials and Methods:60healthy SD rats, cleaning degree, the number of male and female is equal, and theweight is250±20g. The rats randomly assigned into5group, such as normal group, modelgroup, neural stem group, non neural stem group, and nerve transaction group, using randomnumber table. Every group has12rats. Sciatic nerves injury model was made by forcepsholder method. Randomly choosing rats from3rd-5th group, EMG detection was made onthem, to test the MCV. When MVC is below10m/s, sciatic nerves’ axonal and medullarysheath is fractured or seriously injured. According to this, it is sure that nerve injury model ismade successfully. Normal group was feeding regularly, and model group was not. Treatmentwas given for14days at9:00am to neural stem group, non neural stem group, and nervetransaction group after3days. Then AP latency and amplitude and MCV were tested throughgetting electroneurographic signals. And c-fos content in sciatic nerves and around tissueswas tested by immunohistochemical method. Statistics analysis was made on those data.Results:After treatment, the recovery of molding rats’ hind legs was observed. The condition ofneural stem group was improving obviously. Its function of stretching toes and standingreturned to normal basically. The withdrawal reflection results: Stretch reflection andextrusion reflection were normal basically. Hind legs of molding rats could flex and extend when creeping. The condition of non neural stem group was as below: slightly stretching toes;standing, stretch reflection, extrusion reflection and creeping was worse than neural stemgroup. The result indicates that c-fos content of model group in sciatic nerves injury is higherthan blank group, and the difference is significant(P<0.05); The c-fos content of neural stemgroup is lower than non neural stem group, the difference is significant(P<0.05); The c-foscontent of non neural stem group is lower than model group, the difference issignificant(P<0.05); The c-fos content of neural stem group is higher than model group, thedifference is significant(P<0.05). For the electroneurographic signals, comparing modelgroup to blank group, MCV drops obviously, the difference is very significant(P<0.05);comparing non neural stem group to model group, MCV rises obviously, the difference isvery significant(P<0.05); comparing neural stem group to model group, MCV rises obviously,the difference is very significant(P<0.05); comparing non neural stem group to model group,MCV rises obviously, the difference is very significant(P<0.05). MCV of non neural stemgroup and model group rise obviously, but effect of neural stem group is better, the differenceis very significant(P<0.05).Conclusions:after rats’ sciatic nerves injured, acupuncturing neural stem related acupoint--GB30tissues could reduce c-fos content in injured parts. This maybe because the mechanism ofrepairing peripheral nerve through acupuncturing neural stem is nerve axon based oncontinuity. Acupuncturing stimulate switches to nervous impulse, and the nervous impulsemade nurve back to transfer materials, with a slow speed and leaping and continuous way,through chronic demyelination’s axon, and then repairing axon and soma timely, in order tocure compressive peripheral neuropathy. Histology foundation of the poor effect ofacupuncturing non neural stem is that meridians receptors’ pathological damage lead tofunction injury. Thus acupuncturing stimulate cannot switch to nervous impulse. Becauseneural demyelination and conduction block are the chief pathology and function change. So itis inferred that nerve axon’s continuity and acupoint’s receptors’ pathology change may bethat the effect of acupuncturing neural stem to treat peripheral nerve injury is be superior to that of acupuncturing non neural stem. At the same time, considering stimulating differenttissues having different effect on c-fos, clinical efficacy may has difference. In addition, it isfound that the NVC of injured sciatic nerves of acupuncturing neural stem is higher than thatof non neural stem. This explains that acupuncturing neural stem and non neural stem canpromote sciatic nerves’ recovery. This research explain mechanism and curative effect ofacupuncturing different parts, and which has directive function to acupuncturing treatingperipheral nerve injury, and provide scientific theory basis to deep researching and improvingacupuncturing effect.
