Objective To investigate the expression and clinical significance of CD44、CD44v6、TNF-α and TNFR in patients with breast cancer tumor tissue and peripheral blood.Method Draw blood 4ml from 22 cases of breast cancer patients(Associated with lymph node metastasis is 9 cases, 13 cases with no lymph node metastasis), 14 cases of breast benign tumor patients and 15 healthy controls peripheral,Each 2 ml,respectively under contains inert separation gel and coagulant in vitro(yellow) and EDTA anticoagulant tube(purple),Stay in the yellow tube specimens after complete solidification,3000 r/min centrifugal 4 minutes,Using ELISA to detect CD44, CD44v6 and TNF alpha and its receptors TNFR serological levels in separating serum,The purple tube specimens by PCR with fluorescent quantitative method for detecting the level of CD44v6 gene.And to take the malignant tumor and benign tumor group’s breast cancer tissue and its tissue adjacent to carcinoma,After fixing, washing and dehydration, transparent, paraffin, embedding, sectioning, and a series of steps to make paraffin section,And detection the histological level of CD44, CD44v6 and TNFR by the method of fluorescence immunoassay.Result Peripheral blood of breast cancer metastasis in the CD44, CD44v6 and TNF-α and its receptor TNFR were significantly higher than the group without metastasis of breast cancer, benign breast tumor group and the healthy control group, the difference was statistically significant,(P <0.05).The group without metastasis of breast cancer were also significantly higher than those of benign breast tumor group and the healthy control group, the difference was statistically significant,(P<0.05).Which benign breast tumor patients and healthy controls in the peripheral blood CD44, CD44v6 and TNF-α and its receptors TNFR, the difference was not statistically significant,(P> 0.05).Breast cancer breast cancer metastasis in the CD44,CD44v6 and positive expression of TNFR protein content was significantly higher than non-metastatic breast cancer group, benign breast tumor group and the healthy control group, the difference was statistically significant,(P <0.05).The positive expression rate of breast cancer without metastasis group was significantly higher than benign breast tumor group and the healthy control group, the difference was statistically significant,(P <0.05).Which the positive expression of benign breast tumor group, although to some extent, the positive expression rate higher than in normal breast tissue, but the difference was not statistically significant,(P> 0.05).In real-time fluorescence quantitative PCR,peripheral blood of breast cancer metastasis genes CD44v6 levels were significantly higher than non-metastatic breast cancergroup, benign breast tumor group and the healthy control group,the difference was statistically significant,(P <0.05);No metastasis of breast cancer was significantly higher than benign breast tumor group and the healthy control group,the difference was statistically significant,(P <0.05);And benign breast tumor genes in peripheral blood, although slightly higher than the level of CD44v6 healthy control group, but not statistically significant,(P >0.05).Conclusion CD44, CD44v6, TNF- α and TNFR expression levels in peripheral blood and tissue breast cancer group were significantly higher than in benign breast tumor group and the healthy control group,and there is a higher level than those without distant metastasis level transfer.CD44, CD44v6, TNF- α and TNFR prognosis for clinical diagnosis and detect whether the distal metastasis of breast cancer have a certain significance.
