Part I Clinical Significance of Expression of CD44v6 and LYVE-1 in epithelial ovarian tumor【ABSTRACT】Objective The study was aimed at determining the expression of CD44v6 and LYVE-1 in epithelial ovarian tumor and its clinical significance. Methods The expression of CD44v6 and LYVE-1 was examined by immunohistochemical streptravidin-biothin peroxidase(SP) assay. The correlation between their expression and the clinicopathological features was analyzed. Results The expression of CD44v6 and LYVE-1 increased significantly in ovarian epithelial cancer compared to that in benign ovarian epithelial tumors and normal ovarian tissues. The expression of CD44v6 was positively associated with the FIGO stages 1and WHO grades of the patients with ovarian epithelial cancer. The expression of LYVE-1 was significantly related to the stages. The expression of CD44v6 and LYVE-1 was significantly associated with lymph node status.There was statistically significant correlation between CD44v6 and LYVE-1. Conclusions CD44v6 and LYVE-1 played certain roles in oncogenesis, progression and lymphatic metastasis of ovarian cancer. Part II Effect of Estrogen and Progesterone on The Expression of CD44v6mRNA in Breast Cancer Cell Line【ABSTRACT】Objective To study the effect of 17β-estradiol(17β-E2)and medroxyprogesterone acetate(MPA)on the expression of CD44v6 mRNA in breast cancer cell line in vitro. Methods Human brest cancer cell line MCF-7 cells were exposed to 17β-E2(10-12-10-8mol/l)or MPA(10-9-10-5mol/l),the expression level of CD44v6 mRNA was determined by RT-PCR. Results The expression of CD44v6 mRNA in the 17β-E2-treated cells was significantly higher than in the untreated control(P<0.05),the expression leve of CD44v6 mRNA increased with the increase in 17β-E2 concentration. The expression of CD44v6 mRNA in MCF-7 cells treated with MPA(10-9-10-5mol/l) was significantly lower than in the untreated control(P<0.05). Conclusions Estrogen can up-regulate, while progesterone can down-regulate mRNA expression of CD44v6.Further studies are needed to elucidate the mechanism.
