Objective To search for a proper decellular ization method for aortic root homograft, the alterations of biological and immunological properties of rabbit aortic root homograft treated with de-endothelialization and decellularization methods were investigated in the research.Methods Aortic root homografts obtained from rabbits were randomly divided into fresh group, de-endothelializat ion group, and decellular izat ion group. The morphology of homografts was evaluated by hematoxylin-eosin, Masson, VG stainings and scanning electron microscopy. The biological properties such as thickness, tensile strength, broken extension rate, and heat shrinkage temperature were detected. The immunological features(inflammatory cell inf iltration, calcium content and CD152 expression) were determined with immunohistochemistry and flame atomic absorption spectrometry.Result As demonstrated in morphological observation, de-endothelialization and decellularization methods used attained our desired aim perfectly. Tensile strength and broken extension rate of decellularization group were decreased signif icantly compared with fresh group(P < 0.01), while the difference between de-endothelializat ion group and fresh group was not significant(P > 0.05). After allogenic transplantation, inf lammatory reaction was the most severe in fresh group, showing a large amount of inflammatory cells infiltrat ion; compared with fresh group, inflammatory reaction was signif icantly attenuated in de-endothelializat ion group, number of inflammatory cells was significant ly decreased. The inflammatory reaction was the lowest in the decellularizat ion group, only inflammatory cells infiltrated the tissue edge. CD152 expression was significantly increased in de-endothelializat ion and decellularization groups compared with fresh group(P < 0.05). Growth of calcium content in de-endothelializat ion and decellularizat ion groups was significantly decreased compared with fresh group(P < 0.05).Conclusion It could be concluded that the immunogenicity of de-endothelialized and decellularized aortic homografts is decreased signif icantly, and the de-endothelializat ion method could better maintain the tensile property.
