Study On VDR Gene Polymorphism And Susceptibility To Primary Gout In Chinese Han Population

Objective: To explore gene polymorphism of the rs11568820, rs2228570 and rs1544410 of the VDR gene in Han male population of Chinese coastal area, and thus to investigate the relationship between the gene polymorphism of VDR and gout.Methods: 504 patients with gout and 523 gout-free controls were recruited from the Affiliated Hospital of the Medical College, Qingdao University. Genotyping of VDR rs11568820, rs2228570 and rs1544410 was performed by Taq Man allele discrimination assays. An association analysis was carried out using the χ2 test. A genotype–phenotype analysis was also conducted.Results: The analytic results of demographic and clinical characteristics:(body mass index) BMI, systolic pressure, diastolic pressure, blood glucose, total cholesterol and creatinine levels, uric acid, TG and creatinine levels among gout patients were higher than that of controls(P<0.01), while there were no significant differences of age between two groups(P>0.05). Our results showed that polymorphisms of rs11568820 and rs1544410 in VDR were associated with gout in Chinese Han male population. The A allele of both rs11568820 and rs1544410 was associated with the risk of gout [P=0.012 OR 1.251, 95% CI(1.051-1.490); P=0.006, OR 1.574, 95% CI(1.139-2.175)]. However, there was no statistic significance between rs2228570 and gout(P=0.186). Among the genotype-phenotype analysis in gout patients: for rs1544410, there showed a significantly different distribution among the different genotypes(P=0.011) of age at diagnosis 65-84 years and systolic pressure(P=0.007), while for rs11568820, there showed a significantly different distribution among the different genotypes(P=0.011) of disease duration(P=0.039) and creatinine level(P=0.001).We found no statistical difference on the other phenotype characteristics among the three groups.Conclusion: Our study suggested that the polymorphisms of VDR may be relevant host susceptibility factors for the development of gout in Chinese Han male population. However, further study should be done in a larger size sample and other ethic to test and verify our result.
