Objective: Gout is a crystal-related disease caused by deposition of monosodium urate,which is closely related to hyperuricemia.Clinically,gout and hyperuricemia have many influencing factors,among which genes play an important role in their development.ATP-binding cassette transporter G2(ABCG2)can mediate uric acid excretion,which can affect the development of hyperuricemia and gout when genetic mutation occurs.In this paper,two locus of the ABCG2 gene,rs2231142 and rs72552713,were selected,and some male populations in Hebei area were selected as research objects to investigate whether there was a correlation between gout,hyperuricemia and ABCG2 gene and the degree of their correlation,in order to seek the pathogenesis of gout and hyperuricemia from genetics.Method:A case-control study was used in this study.128 male gout patients,132 male hyperuricemia,and 88 male health checkers were selected from the patients in our hospital from November 2017 to November 2018,divided into three groups: gout group,hyperuricemia group and control group.Collecting every patient's clinical data and laboratory test results and their venous blood for biochemical testing and genetic testing.This genotyping method was performed by Snapshot detection method,with 10 samples selected randomly for sequencing verification.All data for this study were statistically analyzed using SPSS 21.0 software.Result:For the ABCG2 rs2231142,there was a statistically significant difference in gene composition between the hyperuricemia group and the control group(P<0.001),of which AA genotype OR=7.50,95% CI=2.47~22.82,AC genotype OR= 2.36,95% CI=1.29~4.34,A gene OR=3.10,95%CI=1.76~5.46;the gout group also had a difference in gene composition compared with the control group(P<0.001),of which AA genotype OR=27.50,95% CI=8.90~84.94,AC genotype OR=6.25,95%CI=3.20~12.21,A gene OR=9.29,95%CI=4.94~17.45.For the ABCG2 rs72552713,compared with the control group,the gene composition of the hyperuricemia group was different(P=0.014),the AG genotype OR=8.70,95% CI=1.11~68.16,the A gene OR=8.70,95% CI =1.11~68.16;however,the difference between the gout group and the control group was not found(P=0.245).The gene distribution of ABCG2 rs2231142 in gout group and hyperuricemia group was different(P<0.001),and rs72552713 showed no difference in gene distribution between the two groups(P=0.162).In addition,rs2231142 single allele mutation was not found to be associated with increased SUA,both allele mutations were associated with increased SUA(P<0.001),and rs72552713 single allele mutation was associated with increased SUA(P=0.005).Conclusion:ABCG2 rs2231142 and rs72552713 are associated with hyperuricemia and gout,and the susceptibility genes of them,and rs2231142 has different gene distribution in gout and hyperuricemia.
