Objective: To investigate the expression and clinical significance of micro RNA-130a/WNT10 A in papillary thyroid carcinoma(PTC) and Hashimoto’s thyroiditis associated with papillary thyroid carcinoma(HT-PTC).Methods: The relative expression of micro RNA-130a/WNT10 A was determined by real time-polymerase chain reaction(RT-PCR) in 41 fresh thyroid tissues(18 PTC tissues, 12 HT with PTC tissues and 11 nodular goiter tissues). The relative expression of Wnt10 a protein was determined by immunohistochemical SP method in 50 paraffin- embedded thyroid tissues(20 PTC tissues, 20 HT with PTC tissues and 10 nodular goiter tissues). The relative expression of Wnt10 a protein was determined by Western Blot in 18 fresh thyroid tissues(6 PTC tissues, 6 HT with PTC tissues and 6 nodular goiter tissues).Results:① RT-q PCR: The relative express levels of micro RNA-130 a in PTCs and HT with PTCs showed a statistically significant difference from those in nodular goiterwith fold changes down to 0.37 and 0.58 respectively(P<0.05).The relative express levels of WNT10 A m RNA in PTCs and HT with PTCs showed a statistically significant difference from those in nodular goiter with fold changes up to 9.98 and 2.69 respectively(P<0.05). ②SP immunohistochemical staining: The expression of wnt10 a protein was obviously higher in specimens of PTC and HT with PTC than in nodular goiter(P<0.05). ③Western Blot: The relative express levels of wnt10 a protein in PTCs and HT with PTCs showed a statistically significant difference from those in nodular goiter with fold changes up to 4.72 and 1.47 respectively(P<0.05).Conclusions: The expression of microRNA-130 a was down-regulated in thyroid carcinoma, and the m RNA and protein expression of its predicted target gene wnt10 a were up-regulated. We predicted that micro RNA-130 a involve in the pathogenesis of HT with PTC by regulating the expression of WNT10 A. The micro RNA-130 had lower expression level in poor prognosis PTC, suggesting that micro RNA-130 a involved in the occurrence of various types of thyroid carcinoma and pathological typing.WNT10 A m RNA and protein showed high expression in thyroid cancer and had difference in pathological typing.
