Objective:By observing the rat model of sequelae of Pelvic Inflammatory suppository for SPID Content of Serum cytokines IL-6 and TNF-a, suPAR impact,Explore its therapeutic mechanism on impact of Action of the SPID,And through Acute Toxicity test of rat and rabbit rectum stimulating test, to evaluate Pelvic Inflammatory suppository clinical drug safety, and provide experimental data for clinical application.Method:1、Study on the mechanism of action:Select 90 healthy female SD rats weight-180g-220g as subjects,Randomly divided into normal group, Sham Operation Group and model group,Apply phenol glue method,On the 14th day give the uterine tissue sampling modeling the evaluation,After the successful model,randomly divided into High、medium and low group of Penyankangshuan and Kangfuxiaoyanshuan group (10 each)、model group (12),Along with the normal group and control group were divided into 7 groups,For 21 consecutive days of rectal application,To observe the morphological changes of rat uterus tissue,ELISA were measured in serum IL-6,TNF-a,suPAR content.2、Toxicological studies:20 healthy SD rats (half male,half female),Randomly divide them into control group and treatment group,apply maximum dose Rectal suspension of Penyankangshuan in rats (6.5625g /ml),For 14 consecutive days,Observation on effect of Penyankangshuan on rat body weight and food intake;Dividel2 healthy rabbits were randomly divided into control group,excipients group and Penyankangshuan group,For 7 consecutive days,Rectum mucous membrane irritation in Rabbits.Result:1、21 consecutive days after drug intervention, HE stain under light microscope to observe the morphological changes of rat uterus tissue, according to the histopathological grading criteria of the uterus, Comparison Between the Normal group and Sham Operation Group, no significant difference (P>0.05); Compared with model control group,normal group and Sham Operation Group, the difference was statistically significant (P<0.01).2、21 consecutive days after drug intervention, HE stain under light microscope to observe the morphological changes of rat uterus tissue, according to the histopathological grading criteria of the uterus,Compared with model control group,Groups with different dosage of Penyankangshuan and Kangfuxiaoyanshuan group.the difference was statistically significant (P<0.01),Compared with low group of Penyankangshuan,high group of Penyankangshuan and Kangfuxiaoyanshuan group,the difference was statistically significant (P<0.05),Inflammatory effect of high group of Penyankangshuan and Kangfuxiaoyanshuan group were significantly better than the high group of Penyankangshuan.3、21 consecutive days after drug intervention,By ELISA were measured concentrations of serum IL-6, TNF-a, suPAR:(1) serum IL-6 concentrations compared with model control group,high、middle groups of Penyankangshuan and Kangfuxiaoyanshuan group,the difference was statistically significant (P<0.05).Comparison Between the low group of Penyankangshuan and model control group, no significant difference (P>0.05),The low group of Penyankangshuan effect of reducing the concentration of serum IL-6 in rats is not significant.(2)serum TNF-a concentrations compared with model control group,high、 middle groups of Penyankangshuan and Kangfuxiaoyanshuan group,the difference was statistically significant (P<0.01),Comparison Between the low group of Penyankangshuan and model control group,the difference was statistically significant (P<0.05),Comparison Between the high and low group of Penyankangshuan,the difference was statistically significant (P<0.05),High dose effects in reducing concentrations of TNF-a in serum of rats better than the low doses group.(3)serum suPAR concentrations compared with model control group,high、middle groups of Penyankangshuan and Kangfuxiaoyanshuan group,the difference was statistically significant (P<0.01),Comparison Between the low group of Penyankangshuan and model control group,the difference was statistically significant (P<0.05).Conclusion:1、By visualizing HE staining and light microscope confirmed histopathological grading scoring of the uterus,By phenol glue molding method SPID can be successfully duplicated model of rat and the procedure has no effect on the model,Is a SPID methods worthy of promotion.2、On the histomorphology observation, By visualizing HE staining and light microscope confirmed histopathological grading scoring of the uterus,Penyankangshuan can relieve the rat endometrium interstitial cell congestion, edema and infiltration of inflammatory cells,Reduce uterine cavity expansion to fix improve pelvic pathology.3、Judging from the results of the experimental index expression,Penyankangshuan treat SPID may inhibit the production and release of serum IL-6、TNF-α suPAR in rat model,Reducing the body’s inflammatory response and injury, and promote the recovery of the Organization,Inhibition of Pathologic development of the SPID, play a role in treatment,Therefore, we can think of inflammation-related cytokines IL-6、TNF-α、suPAR therapeutic targets for Penyankangshuan treatment.4、Through acute toxicity studies in rats and rabbits rectal mucosa irritation test results suggested,No acute toxicity and rectal Penyankangshuan rectal mucosa irritation, clinical medication safety is high, and is worth popularizing.
