Objective:(1) to observe the penyankangshuan anti-inflammatory and analgesic effects of xylene induced ear swelling in mice; plastic ring induced chronic granulation swollen,acetic acid induced writhing in mice. (2) by observing the changes of inflammatory cytokines in serum of rats with chronic pelvic inflammation in the discussion of pen Yan Kang suppository in the treatment of sequelae of pelvic inflammatory disease (sPID) and the mechanism of anti-inflammatory action.Methods:(1) on mouse ear swelling induced by xylene, calculate the degree of swelling, observe the influence of penyankangshuan on acute inflammation. Plastic ring to chronic granulomatous model, to observe the effects of penyankangshuan on chronic inflammation. Using acetic acid in mice body torsion experimental observation penyankangshuan analgesic action. (2) Using phenol mucilage agent in rats caused by chronic endometritis to represent the chronic pelvic inflammation model in rats, Respectively set the high dose group of penyankangshuan (equivalent to 2.52g crude drug/kg), middle dose group penyankangshuan (equivalent to 1.26g crude drug/kg), low dose group of penyankangshuan (equivalent to 0.42g crude drug /kg), Kangfu anti-inflammatory suppository group, model control group, sham operation group and normal control group.15 days after modeling, continuous enema administration for 21 days, ELLISA was used to detect the serum SDF-1, TGF-beta 1 and IL-10 level;the naked eye and optical microscope HE staining to observe the pathological changes of rats uterus.Results:(1) penyankangshuan high, medium and low dose group compared with the blank group can reduce the degree of mouse ear swelling caused by dimethylbenzene, high, medium and low dose group compared with the blank group was extremely significant difference (P<0.01).(2) penyankangshuan high and medium dose group compared with the blank group, were able to reduce the weight of granuloma, there is a very significant difference (P< 0.01), penyankangshuan low dose group compared with the blank group there was no significant difference(P>0.05).(3)Compared with the normal control group, control group of rats uterus tissue HE staining positive score have no difference (P> 0.05),model group rats palace HE staining positive score was statistically significant (P< 0.01),description: pelvic inflammatory disease sequelae model building success. Compared with model group, penyankangshuan low, medium and high dose of rat uterine histopathologic grading were highly significant (P<0.01).(4)Pelvic inflammatory disease sequelae model rats serum anti-inflammatory cytokine IL-10 levels compared with normal control group rats decreased, TGF-beta 1, the SDF-1 expression level was higher than normal control group, were extremely significant difference (P< 0.01). After drug intervention, penyankangshuaneach dose group of rats serum TGF-beta 1, the SDF-1 level than model group decreased, anti-inflammatory cytokine IL-10 levels were higher in the model group.Conclusion:the above results, (1) penyankangshuan has control of acute and chronic inflammation and analgesia effect; (2) penyankangshuan can obviously reduce the model rats of chronic inflammation of the lining damage, reduce the excessive inflammatory cells infiltration, reduce the proliferation of fibrous tissue. (3)penyankangshuan for chronic endometritis endometrium of rats with good anti-inflammatory effects, May be by inhibiting model rats serum SDF-1, TGF-beta 1 activity, raised the IL-10 levels, and And reduce the excessive inflammatory cells infiltration and formation of inflammatory mediators, reduce the fibrinogen, to increase the degradation of hyperplasia of fibrous tissue, promote local blood circulation, to repair the diseased tissue, and reduce inflammation reaction through the multiple channels.
