Anemarrhena asphodeloides Bge. belongs to the family Liliaceae widely distributed in China. Its rhizome has been used as a folk medicine for treatment of febrile diseases with high fever and dire thirst; heat in the lung with dry cough; consumptive fever; diabetes due to internal heat; constipation in China. It has been reported that steroidal saponins were the main active components. In these years, there are so many articles to report about the timosaponin and sarsarsapogenin.The author has investigated the chemical constituents,synthesized sarsasapogenin derivatives,and then evaluated their antiaging activities of the Rhizoma anemarrhenae.In this thesis,11 compounds were isolated with silica gel,ODS,Sephadex LH-20 and preparation HPLC, etc. On the basis of analysis of UV,NMR,MS data and measuring of physic-chemical characters, all of the chemical structures compounds were elucidated as timosaponin AⅣ(1),timosaponin AⅢ(2),sarsasagogenin (3),timosaponin BⅡ(4), timosaponin B I (5), timosaponin B V (6), timosaponin BⅢ(7),macrostemonoside F (8), marcogenin (9),β-sitosterol (10), daucosterol (11). of all these compounds.In order to explore the relationship of structure and activities and find some compounds with good pharmacological activities, we have isolated the sarsasapogenin.By means of extracting,hydrolysis and recrystallization, on the basis of chemical and spectral analyses,its structure and purity were identified.We have done the structure modification of sarsasapogenin isolated, and synthesized 11 sarsasapogenin derivatives. (6 of them were new compounds)The isolated compounds and sarsasapogenin derivatives were examined for their effects on the antiaging activity from human umbilical vein endothelial cell.All of them have different kinds of antiaging activities.The sulfated sarsasapogenin dervative and timosaponin BⅡhave the significantly good activities and low toxicity.
