Pleurotus eryngii is a kind of edible and medicinal rare edible fungi which was manually cultivated successfully in recent years. Presently, many researches on relevant aspects of Pleurotus eryngii were reported. But more attention had been focused on optimum formula cultivated material and cultivation technique. Study on excellent strains selection in Pleurotus eryngii was rarely reported, which is lagging seriously. In this experiment,47 strains of Pleurotus eryngii collected from different areas of China were used as materials to select excellent parental strains by cultivation, antagonistic test, high temperature resistance test and ISSR analysis. Monospore crossbreeding and protoplast monokaryon crossbreeding were used for study on excellent strains selection in Pleurotus eryngii.In the cultivation of 47 strains of Pleurotus eryngii, fruitbodies were not successfully produced by 11 strains. In the 36 producing strains, biological efficiency of 14 strains were higher than 50%, and that of strain pl.e06 was 73.51% as the highest. The 14 strains were divided 7 categories though antagonistic test. For each category, the one which had highest yield was selected as primary parental strains, they were strains pl.e03, pl.e06, pl.e10, pl.e13, pl.e16, pl.e24 and pl.e29. The 7 primary parental strains were cultivated repeatedly, used for high temperature resistance test and ISSR analysis. The results showed that strains pl.e03, pl.e06 and pl.e16 had better agronomic characteristics; pl.e16 had highest temperature resistance; the genetic similarity coefficient between pl.e06 and pl.e16 was 0.4503 as the smallest. On the basis of above results, pl.e06 and pl.e16 were selected as parental strains.Monospore crossbreeding and protoplast monokaryon crossbreeding were used to get 65 and 37 hybrids respectively. Growth rate of hybrid isolates were tested in the CYM medium and compost.13 hybrids from monospore crossbreeding and 2 from protoplast monokaryon crossbreeding were preliminarily selected for high temperature resistance test. It was found that 15 tested hybrids were inferior to parental strain pl.e16; D20, D42 and D48 from monospore crossbreeding had higher temperature resistance relatively. The 15 hybrids were cultivated and the grey correlation analysis was conducted for a comprehensive evaluation based on 7 selected traits including pileus diameter and thickness, stipe length and diameter, average fresh weight, biological efficiency, period from inoculation to primordium formation. The results showed that the correlation degrees of D12, D17, D18, D19, D20, D30, D31 and D48 from monospore crossbreeding ranked ahead of 2 parental strains with increased agronomic characteristics; the degrees of Y29 and Y30 from protoplast monokaryon crossbreeding ranked between the parental strains.
