In this paper, a high-performance liquid chromatography was developed for simultaneous determination of florfenicol and florfenicol amine in plasma and tissues of yellow catfish(Pelteobagrus fulvidraco). And a pharmacokinetics and tissues residue depletion was studied by this method at 25℃water temperatures in healthy yellow catfish after single does oral administration and multidoes oral administration.1. A high-performance liquid chromatography was developed for simultaneous determination of florfenicol and florfenicol amine in tissues of yellow catfish. The extraction solvent is acetone, and the residue is defatted with n-hexane. The HPLC system used was the Agilent 1200 series equipment, detection of florfenicol was performed on the variable wavelength detector at 223nm, the column temperature is 30℃and and the flow rate was 1.0mL/min. The consist of of mobile phase is acetonitrile-sodium phosphate buffer (30/70,V/V) solution (pH=3.8), and the flow rate was 1.0mL/min. The result showed that the stanbdard curves were linear for two analytes at 0.05~10.00 ug/mL of florfenicol and florfenicol amine. The average recovery of florfenicol and florfenicol amine in tissues was 81.4%-89.0% and 80.4%-85.7%. The detection limits for florfenicol and florfenicol amine were 0.025μg/mL and 0.015μg/mL in plasma, respectively, were 0.015μg/g and 0.010μg/g in muscle, were 0.030μg/g and 0.030μg/g in skin, were 0.025μg/g and 0.025μg/g in liver, were 0.050μg/g and 0.025μg/g in kidney. Consequently, this method can be used to detect simultaneously florfenicol and florfenicol amine residues in yellow catfish.2. Pharmacokinetics and tissue distribution of florfenicol in yellow catfish at 25℃were studied after they were given a single oral dose of 10 mg/(kg.bw). The reslts shows that plasma concentration-time curves, muscule concentration-time curves and skin concentration-time curves were best described by a one-compartment model with 1st order absorption, kidney concentration-time curves and liver concentration-time curves were best described by a two-compartment model with 1st order absorption. The distribution volume (Vd/F) of FF was computed as 2.52L/kg. In tissues, Time to maximum concentratiotn in the order of magnitude were liver(1.82h)
