In present study we use long photoperiod-sensitive genic male-sterile rice N5088S,NW04S and short photoperiod induced male-sterile rice D52S to identify the proteome changes in young panicles after LD treatment(14.5h) and SD treatment (10h). The various photoperiodic treatments are from the differentiation of secondary branch stage to the stage of meiotic division of pollen mother cell in the same temperature. Effects on pollen fertility after treated in SD and LD conditions are obvious.Comparative analysis of young panicle proteome under sterile and fertile conditions are made by two-dimensional gel electrophoresis. Identified proteins that differentially expressed are further characterized by MS/MS. The main results are as follows:1. Effects on pollen fertility after treated with various photoperiodic treatments are obvious. As compared with D52S treated with 10h SD during photoperiod-sensitive stage,14.5h LD treatment significantly increased the fertility and as high as 54.3% fertility was observed. Treated with 14.5h LD, pollen from N5088S and NW04S are completely sterile while 10h SD treatment increased the fertility to 81.0% and 74.8% respectively.2. Comparative studies on proteins from the young panicle of D52S, N5088Sand NW04S under sterile and fertile conditions from the formation stage of pollen mother cell to the stage of meiotic division of pollen mother cell are made by two-dimensional gel electrophoresis. We obtain satisfied 2-DE maps.Twenty-eight proteins are further characterized by MS/MS and fourteen proteins are successfully identified. In D52S, retrotransposon protein is specially expressed under fertile conditions while adenosine kinase is occurred only under sterile temperature. The up-regulated proteins under sterile temperature are Sucrose synthase, Eukaryotic translation initiation factor 3 subunit K and may be other proteins like ATP synthase CF1 alpha chain, Alcohol dehydrogenase 1 and calcium-dependent protein kinase. The expression of small GTP-binding protein and phytochrome-like ATPases are stimulated in fertile conditions. In NW04S, P-loop containing Nucleoside Triphosphate Hydrolases and Aminoacyl-tRNA synthetase are specially expressed under sterile conditions. The up-regulated proteins under sterile temperature are nucleotidyl transferase superfamily, Eukaryotic translation initiation factor 3 and may be other proteins like Catalytic domain of Protein Kinases,Histidine kinase-like ATPases, calcium binding motif, retrotransposon protein, Ty3-gypsysub-class and Glycosyl hydrolases. The expression of Protein serine/threonine kinase is stimulated in fertile conditions. In N5088S, pepsin-like aspartate proteases and Putative methionyl-tRNA synthetase are specially expressed under fertile conditions. The up-regulated proteins under fertile temperature are Short-chain dehydrogenase/reductase SDR and may be other proteins like Chromosome segregation ATPases, Zinc knuckle,Phosphofructo-kinase and Tat binding protein 1(TBP-1)-interacting protein. The expression of P-loop containing Nucleoside Triphosphat is stimulated in fertile conditions.3. Compare with those two sorts of genic male-sterile rice, the proteins identified in Glucose metabolism and Energy metabolism show some common under sterile conditions.The large number of differentially expressed proteins we gained in this study provides a basic data to further explore the mechanism of photoperiod-sensitive genic male-sterile rice.
