Lily which belongs to perennial flower bulbs is one of the most important fresh cut flowers around the world. There is extremely high economic value and ornamental value for the species. From the current situation of lily breeding, much of the work aim to improve the ornamental characteristics. But the resistance breeding has rarely been reported. In this study, two aspects including salt-tolerant evaluation and EMS mutation in vitro of lily were investigated. The results as follows:1.Through micropropagation of six cultivars and two wild species, we got large numbers of in vitro plantlets, and then using MS medium containing different concentrations of NaCl to screen the plantlets.According to the performance in different salt concentrations,salt-tolerant threshold for different cultivarts were determined, which is 14g/L for'Manissa',13g/L for'Bernini',12g/L for'Co Amore' and'Acapulco',and lOg/L for'Simplon',Wild lily in Yunnan, Easter lily and 'Siberia'.2.Put the micropropagated plantlets in MS medium with a same salt concentration, random sampled and then measured physiological index. According to the MDA content and the activity change of SOD, POD and CAT, the degree of salt tolerance can be evaluated. According to comprehensive evaluation, the strength of salt tolerance of different cultivars was'Manissa'>'Bernini'>'CoAmore'>'Acapulco' >'Simplon'>wild lily in Yunnan>Easter lily>'Siberia'.3.The salt-tolerant threshold of Easter lily is 10g/L NaCl according to the growth of callus into the medium containing different salt concentrations.4. The calli were treated with different concentrations of EMS solutions for different periods. The results indicated that low concentration of EMS solutions can improve the survival rate of callus while high concentrations suppress the growth. When the EMS solution is at the same concentration, the longer time is, the lower callus survival rate is. The optimal mutation dose for Easter lily callus is 0.6%+2h.5.Salt tolerance enhanced plants were differentiated from the calli treated with EMS.Through physiological index measurement, the results showed MDA content of these plants was always lower than that of control, while the activity of SOD and POD were always higher than that of control.The CAT activity after salt stress for 9d is higher than that of control.
