Papaya is a kind of valuable Chinese herb, with high food value and nutritional health protection effects, but fresh Papaya fruit is seldom eaten because of its acerb taste, there are mainly papaya wine, fruit vinegar, preserved fruit , candied fruit and other traditional foods at present. The utilization is not high。This paper study on the best process of extraction, separation and purification of physiologically active compounds in Papaya. And we also studied the extraction and separation of the papain and SOD of papaya with activity has researched, which provided a theoretical basis for the comprehensive utilization of papaya. This study has a great significance on improving the economic benefits for the majority of farmers, expanding OA, papain and drug source of SOD, meanwhile, it aslo has a great importance on promoting the development of Anhui papaya industry.1 The nutrient component in fruits of Xuancheng Pawpaw was determined, it contained total suga2.62%,diminishing sugar1.35%,organic acids 2.42%, pectin 0.89 %, tannin 2.21%,Crude fiber 1.12%, protein 1.36 %, Ursolic Acid 0.11 % ,OA 0.20%, Anti-oxidative0.15, the activity unit of papain was 600.72 U; the activity of SOD was 1460.00U.2 Then extracted through such approaches as refluxing extraction,microwave assisted extraction,ultrasonic wave assisted extraction, Infusion extraction and Soxhlet extraction. Such approaches mentioned above were optimized, as a result,the ultrasonic wave assisted extraction was better extraction technique, the parameter as follows: 95% ethanol as extraction solvent, extraction time for 10min, microwave power 200W, extraction once.3 Based on the same group of extract separately spectrophotometry and HPLC testing, the results show that, samples of spectrophotometry easily influnced by other color, high impact assessment, HPLC isn`t influnced by other impurity ,and has high precision.4 To establish a HPLC method for the determination of oleanolic acid in Xuan Papaya. A Phenomenex C18 Column (250mm×4.6mm i.d., 5μm) was used with the mobile phase being methanol-(pH=2) aqueous solution of phosphoric acid (85:15), flow rate being 0.8mL/min,determinating wavelength being 210nm,the column temperature being 27℃. The simple method , high precision and good repeatability provide a reliable scientific basis for Xuan Papaya in the determination of oleanolic acid. 5 Acid deposition method, chloroform extraction and macroporous resin chromatography separation were used respectively for seprating and purificating OA, the purity of the objects from the three methods were: 41.02%, 57.08% and 70.56%.6 The activity of papain and SOD which were from crude papaya enzyme solution were respectively measured: The activity of papain was 600.72U.7 The activity of SOD was 1460.00U, specific activity was 2.08U/mg.
