Chaenomeles speciosa Nakai Fruit contains a variety of nutrients needed by the human body, is a kind of medicine food dual purpose type of fruit, main purpose is medicinal. This paper aims to study speciosa oleanolic acid and ursolic acid extraction, separation and purification process optimization, establish the speciosa GC-MS chromatographic fingerprint based organic acids for the characteristic ingredients, the results has important theoretical and practical value for development and utilization the Speciosa resource and quality control of the Speciosa herbs. This thesis with Speciosa as experimental materials, organic acids as the research object, were studied oleanolic acid ursolic acid extraction and purification process optimization by HPCPC separation and HPLC detection technology; the organic acids from12samples of authentic region of the country’s material are analysed by GC-MS, established GC-MS chromatographic fingerprint of organic acids, and Speciosa fruit from Qijiang in Chongqing were analyzed the components of organic acids. The research results are as follows:Optimized oleanolic acid and ursolic acid extraction process, the regression equation that it expresses the relationship between the total extraction rate and the influence factors and the HPCPC separation conditions of oleanolic acid and ursolic acid was established. The results showed that oleanolic acid and ursolic optimum extraction process of extraction time was40min,46℃of extraction temperature,43mL/g of solid-liquid ratio,88%of ethanol concentration; The HPCPC separation conditions of oleanolic acid and ursolic: solvent system of n-hexane-ethyl acetate-methanol-water (7:3:5:1, V/V), flow rate1.5mL/min, speed1000rpm, the purity of oleanolic acid and ursolic acid96.9%and93.4%under this condition.The organic acids from12samples of authentic region of the country’s material were analysed by GC-MS, established GC-MS chromatographic fingerprint of organic acids by abstracting common peak, calculating chromatogram similarity, determines21organic acids common chromatographic peak. Speciosa fruit organic acids from Qijiang in Chongqing were analyzed by GC-MS,37compounds were identified by calculating the relative content of each component according to the area normalization method, by mass spectrum database retrieval and reference literatures.
