1BL·1RS translocation lines played an important role in wheat breeding and production in China. Rapid and accurate identification of 1BL·1RS translocation is important for wheat improvement. In order to discriminate the 1BL·1RS translocations from different origins, 10 STS, one SCAR and four RAPD markers specific to 1BL·1RS translocations as well as 22 SSR markers located on chromosome 1RS were employed to test 78 accessions with 1BL·1RS translocations, 10 non-1BL·1RS translocations, and three rye lines. The 1BL·1RS translocation lines included Zhou 8425B and its 36 derivatives, Lankao 906 and its five derivatives, Aimengniu and its eight derivatives, and Lovrin series with 1BL·1RS translocation (29 accessions). Four STS, one SCAR and five SSR markers for different stem rust resistance genes were employed to test resistance genes in 308 wheat genotypes accessions, including 32 CIMMYT lines and 276 Chinese materials. The results are summarized below.1. Seven STS, one SCAR, three RAPD, and three SSR markers can be effectively used to identify the 1BL·1RS translocations. Among them, the markersω-sec-p1/ω-sec-p2,ω-sec-p3/ω-sec-p4, H20, and SECA2/SECA3 were better, with stable amplification, clear bands on electrophoresis gel, good repeatability, and simple operation.2. In the 78 lines with 1BL·1RS translocations, no genetic diversities were observed in the 1RS translocations from different origins in the test with molecular markers.3. AF1/AF4 and Iag95 markers can be used to identify originally the stem rust resistance gene Sr31, Gb marker can be used to identify originally the stem rust resistance gene Sr25, gwm389, gwm533 and barc133 can be used to identify originally the stem rust resistance gene Sr2. Of them, the markers AF1/AF4, Iag95, Gb, and gwm 533 were better, with stable amplification, clear bands on electrophoresis gel, good repeatability, and simple operation.4. Among 308 wheat accession, Sr31 is present in 67 varieties, Sr25 is present in 12 varieties, and Sr2 is present in 175 varieties.
