Resveratrol is a kind of polyphenolic compounds, which are featured with anticancer, antibacterial, estrogenic, antitumour, nervous system protective and other pharmacological activities. It widely exists in plants, such as, Arachis Grape, Peanut, Hypogaea and Mulberry. This paper reported on the study of Peanut which is contain abundant resveratrol. We carried out the induction of callus and the suspension culture of Peanut cell in the shake flask, constructed a non-structured kinetic model and studied the effect of elicitors on cell growth and resveratrol accumulation in root cultures of Peanut.Different media, different explants and different hormones combination had significant influence on callus induction of Peanut and resveratrol content. The best hormone combination was determined by the quadratic regression general rotation design with three factors. The optimal medium for the induction of Peanut was MS, combined with 2.5 mg/L 2,4-D,1.8 mg/L KT and 3.0 mg/L 6-BA. The maximum biomass from Peanut cell reached 43.95 g/L, and the high yield of resveratrol reached 9.073 mg/g.In the course of suspension culture, the effects of rotation ratio, inoculum density, pH and sucrose concentration were investigated. The conditions of suspension cell culture were determined by the quadratic regression general rotation design with four factors. It was as follows:130 r/min,38 g/L inoculum,5.8 pH and 40 g/L sucrose. The determination of the protein content, phenylalanine ammonialyase activity and peroxidase activity react to the growth of suspension culture cell and the changes of resveratrol content.A non-structure kinetic model of the growth of Peanut cell suspension was constructed. This model described the rule of Peanut root cell growth, limiting substance consumption and resveratrol production. These results showed that the model had a very high coincidence with the experimental dates. These equations of kinetics model were as follows respectively,SA had no obvious effects on the biomass accumulation of Peanut root cell. Excess SA could cause cell browning, while low concentration ethrel showed a significant result on the growth of cell (< 5 mg/L). SA and ethrel could promote the accumulation for the increase of resveratrol.5mg/L SA improved the yield of reveratrol slightly after 9 d of elicitation(up to 8 mg/L), while 5 mg/L ethrel had a better effect after the treatment of 9 d (up to 96 mg/L).
