Based on formal research, this paper determined its optimal process condition,and further purification was made with Column Chromatography(CC); the optimal formulation of 10% Radix Peucedani coumarin ME was screened out with single-factor attempting method; Meanwhile, optimal applying time and concentration of 10% Radix Peucedani coumarin ME were conformed through indoor assay and field assay.Results were showed as follows:(1) Single-factor experiment and orthogonal experiment showed the optimal extraction condition was temperature of 60℃, pressure of 30MPa, extraction time of 1.5h, crush degree of 10mesh.(2) With high efficient thin layer chromatography,optimal solvent elution system of SFE extraction was serened out as petroleum ether and ethyl acetate and two components with remarkable characteristics of Radix Peucedani coumarin were separated.(3) The optimal formulation of 10% Radix Peucedani coumarin ME was screened out with single-factor attempting method:coumarin extraction 10%, emulsifier 12%, petroleum ether and ethyl acetate (mass ratiol:1) 4.5%, glycerol 0.6%, sodium sorbate 0.5% and water.(4) Through indoor assay and field assay, screened out that optimal applying time was three-leaf stage and optimal applying concentration was 200×diluent. Control effect of 200×diluent was 80.42% or 81.34% respectively in indoor assay and field assay, which is equivalent with tricyclazole.
