The experiment studied the techniques of test-tube seedlings tissue culture rapid propagation in different varieties of ZJM. The optimal culture medium and conditions for plantlet regeneration had been selected under the variant phytohormones combinations and culture conditions. Moreover, the study discussed various factors, which had effects on ZJM differentiation and growth and raising root culture.The results are as follows: the suitable medium of primary culture was MS medium supplied with 0.5mg/L BA+0.2mg/L IBA in all three different varieties. But they had different result in subculture. Supplements of BA at 0.6mg/L and IBA at 0.2mg/L attached to the MS medium were suitable for shoot differentiation in hu ping ZJM. But Ban ZJM were beneficial for the shoot differentiation in the medium of MS+0.1.mg/L BA+0.5mg/L IAA. Supplements of 2.0mg/L BA+0.5mg/L BA added to the MS medium were good to subculture of jun ZJM. sucrose Concentrations attached to the MS medium were different from each other in shoot differentiation. Huping ZJM were robust in high sucrose concentrations. But ban ZJM were more suitable in low concentrations. Jun ZJM were insignificant effect in sucrose concentrations. The culture temperature, illuminating intensity, illuminating period for differentiation were 20-30 ℃ , 3000~4000Lx, 16h respectively. The different phytohormone had different result on raising root culture in different varieties. With 1/2MS and sucrose 15g, IBA 2.5 mg/L had a good result on raising root culture in huping ZJM, but IAA 2.0 mg/L in ban ZJM and IBA l.0mg/L in jun ZJM were suitable for raising root culture. The best culture conditions was 28~30℃ for the first ten days and 25℃ constant temperature culture for a month subsequently, With illuminating intensity and illuminating period 1000~2000Lx, 16h respectively. The more important techniques of ensuring transplant living rate were: (1) it was necessary to select test-tube seedling of healthy and strong; (2) it was needed light practicing seedlings before transplant; (3) humidity was higher than 90%; (4) the best period for transplant was from May to July.This research set up a succeed techniques guarantee for industrial raising seedling and spreading in the production of ZJM.
