Genetic Diversity Of Piuns Koraiensis Populations From Hei Longjiang Area Revealed By ISSR Analysis

With ISSR method, the paper studied genetic diversity and genetic differentiation of 238 individuals in 13 populations of Piuns koraiensis populations from Hei Longjiang and revealed the genetic relationship and mechanism of genetic differentiation of this area, inquired into the genetic structure and provided the rationale of the effective protection and reasonable use. The researchful results as follows:1,Thirteen ISSR-PCR primers that had vivid amplification band and abundant polymorphism had been screened.Thirteen primers which were screened out with ISSR technique were able to generate 139 loci in total, there are 96 polymorphic loci and the percentage of polymorphic loci is 0. 6906. Nei's index is 0. 2403 , and the Shannon's index is 0. 3598. The result shows that there are more genetic variations in Hei Longjiang province.2,Coefficient of genetic differentiation is 0.3404. Genetic diversity among populations possessed 65.94%of the total and genetic diversity between populations possessed l9.99%. We can conclude that Genetic diversity mostly exit in populations; Nm is 0.9689 which means that the gene flow is nature. However, it is not high. A higher level of genetic differentiation was detected among population. So we can confer that the resource of the Piuns koraiensis was destroyed of mankind,So the habitat was destroyed and the population was fregmentray. Aimed at this situation,we advance the suggestion of the protection of Piuns koraiensis resource.3,Piuns koraiensis populations from Hei Longjiang can be measure off 3 areas of the 13 populations. They are mainly fasten on Xiao Hinggan Ling and Zhang Guangcai ling. We can speculate that natural Piuns koraiensi in China have been differentiated as early as the Quaternary Ice Age occurred, the regions may be the refuge for natural Piuns koraiensis in Xiao Hinggan Ling and Zhang Guangcai ling of Hei Longjiang...
