There is rich resources of Cornus Officinalis in Shitai of Anhui province. Cornus valuable as our traditional Chinese medicine, has a variety of chemical constituents and pharmacological, physiological effects, and it has broad development prospect. To use Cornus Officinalis resources fully effectively, papers on the Anhui region of the basic components of Cornus officinalis measured to study the extraction of ursolic acid from Cornus officinalis, and purification process, also discussed the role of the antioxidant UA.The experimental determination of the Cornus Officinalis included sugar, total sugar, organic acids, nitrogen, crude protein, crude fiber, ursolic acid and oleanolic acid content.The content is 8.362% 16.37%, 6.28%, 0.4324%, 2.7025%, 2.32%, 0.2722%, 0.18% respectively.SDE was used to extract essential oil from Fructus Corni of Shi Tai,. Anhui provience.Qualitative and quantitative analysis were analyzed by GC-MS. SDE was used to extract essential oil from Fructus Corni,it`s a pre-processing method by concurrent heating the sample and organic solvent to boil .The amount of the components from the essential oil were determined by normalization method.The components separated were identified by GC-MS. The results showed essential oil of Fructus Corni could be extracted effectively by this method,48 essential compounds were identified.The relative percentages of the constituents were determined by normalization method.The highest content was hexadecanoic acid(28.86%),followed by Methyl linolenate (10.82%).They composed of 91.54% of the total essential oil.Through TLC on the extract of Cornus Officinalis, qualitative identification and UA found in the same Rf value standard acidity of the same place of purple spot.This paper gives extraction optimization of ursolic acid from fructus cornis. Through orthogonal experiments, the best condition of ultrasonic extraction is as follows: 80% ethanol, solid-liquid ratio 1:20 ,temprature 35℃, supersonic wave treatment for 15 min. The suitable method of removing the impurities is using petroleum ether to elute the extracting solution for three times with 0.2 times volume of the extracting solution, and then using carbon tetrachloride to elute for three times with 0.3 times volume of the extracting solution.Anti-oxidation experiments In vitro:Application of chemical reaction method to detecting the ursolic acid scavenging effect on superoxide anion from pyrogallol auto-oxidation reaction and the hydroxyl radical of the Fenton reaction; Anti-oxidation experiments In vivo: Kunming mouse were randomly divided into blank control group, aging model group, positive control group, low-dose group and high-dose group, 10 in each group. After the administration of 42d, pick eyeball blood,detection the mouse of serum superoxide dismutase (SOD) activity and malondialdehyde (MDA) levels, also detection its thymus index and spleen index. Ursolic acid has a clear hydroxyl radicals and superoxide anion radical role, it can significantly increase the serum superoxide dismutase (SOD) activity and malondialdehyde (MDA) content,besides,thymus index and spleen index of the Ursolic acid groups mice were significantly increased, P<0.05.Conclusions: Ursolic acid has significant antioxidant activity, it provides a basis for the further development of medicinal herbs Cornus.
