Fructus Comi is the pulp of the ripe fruit of Comus officinalis Sieb.Et Zucc. (Comaceae,it is called dogwood fruit in English).It can supplement the liver and kidney, astringe essence and stem desertion Pharmacological research on Fructus Comi has showed that it can regulate immune function of mice and decrease blood sugar in diabetic mice.Ursolic acid belongs to triterpenic acid,it has tranquihzation,anti-inflammation, strength immunity,anti-bacteria,beautiful white and anti-tumor pharmacological actions. Thus much more attention has been paid to the studies of Ursolic acid since it was experimental proved having the hope to become a new potent natural anti-cancer medicine of low-poison and high-validity by the tumor hospital of Chinese acadmy of medical in recent decades.This paper is studying on the process of isolating and purifying Ursolic acid from Comus officinalis Sieb.et Zuec..It detailedly summarize the current research situation of the original plants,development in researches of Ursolic acid in phamlacological actions and the methods of separation and purification.A practical process flow and the optimal process of extracting Ursolic acid from Comus officinalis Sieb.et Zucc.were afforded by doing lots of experiment.Based on the qualitative experiment of Thin Layer Chromatography,Ursolic Acid in fruits of Comus officinalis Sieb.et Zucc.were found.Then extracted through such approaches as Infusion extraction,Soxhlet extraction, refluxing extraction,microwave assisted extraction and ultrasonic wave assisted extraction. Such approaches mentioned above were optimized,as a result,the microwave assisted extraction was the best extraction technique,the parameter as follows:extraction time for 6 min,70%ethanol as extraction solvent,microwave power 300W,ratio of material to liquor 1/12 g/ml,the yield is 10.62%,the purity reaches 15.36%.Ursolic acid was separated and purified by macroreticular resin chromatography separation,and chose the concentration of desorbing solvent.Then,the product is obtained from the concentrate of 80%alcohol eluent(pH=10.0).The results show that 80%alcohol (pH=10.0) as desorbing solvent was best.Finally,the recovery is 86.95%and the purity is 76.64%.A quick method to separate and to test Ursolic acid—High Performance Capillary electrophoresis(HPCE) was stated.The ursolic acid was baseline separated under the optimum buffer condition of 15mM disodium hydrogen phosphate+15raM sodium borate (pH=9.22)+10%carbinol(v/v) at 214 nm of the wavelengh and 20.0 kV of constant voltage.And the content of UA was 0.2171%by HPCE.
