Plant cell culture produce secondary metabolites have many advantages, such as protection of resources,cost savings and controllable conditions,short production cycle, not subject to geographical and time constraints,and conducive to industrialized produc-tion.Secondary metabolites resveratrol in grapes is a stilbene structure of non-flavonoidpolyphenol compounds,because of its antibacterial anti-inflammatory, anti-infective, anti-virus,anti-oxidation,anti-cancer, protect the liver, radiation protection,immunity regulation,protection of cardiovascular and estrogen-like activity of a variety of pharmacological effects and biological activity of Taxol has been hailed after following another new green anti-cancer drugs, has been widely concerned.In this experiment, callus induced from the leaves and stems of Vitis vinifera L"Pinot Noir"were used as materials, by adding precursors (cinnamic acid and phenylalanine) and additives (salicylic acid,colchicine andβ-cyclodextrin) to examine the different adding time and harvest time on the vine callus proliferation and the dynamic synthesis effects of resveratrol. Followed by callus induced from the Pinot Noir leaves as the material for investigating the effects of different light source qualities and LED photoperiods on the growth and resveratrol synthetic dynamic.Finally, Pinot Noir leaves induced callus as starting material to study the grape cell suspension culture growth and the different concentrations of additives on the synthesis of resveratrol in grape suspension cells.The results show that: Under appropriate concentration of different precursors and additives, the 0d adding cinnamic acid, phenylalanine,salicylic acid,colchicine andβ-cyclodextrin,During 31d culture time, harvested callus at different times, appropriate harvesting time for resveratrol content were the 11th, 11th, 1st, 3rd and 1st day, respectively, and yield suitable harvest time were the 23rd, 29th, 1st, 29th and 29th day. Adding above substances after inoculation every 2 days, the 33rd day unified harvest, appropriate adding time for resveratrol content were the 22nd, 32nd, 22nd, 12nd and 10th day, respectively, and the optimum production adding time were the 30th , 32nd, 16th, 12nd and 32nd day.Fluorescence under different light qualities,white light was beneficial to the callus growth, but suppressed the resveratrol synthesis; green light promote resveratrol production increased obviously, followed by blue light; red light significantly increased the content of resveratrol, but go against resveratrol accumulation in callus.In different LED light qualities,GLED was beneficial to the callus growth,but suppressed the resveratrol synthesis.BLED had no significant effect on the cell growth,but it had stronger promotion on content and production of resveratrol,while RLED is not conducive to the accumulation of resveratrol.During 30d culture period,and the 24th day was the ideal harvest and subculture time.Among BLED different photoperiods,8h/d light treatment had significantly inhibition on resveratrol synthesis,and the rest photoperiods had no obvious difference.Added precursors and salicylic acid to the grape cell suspension system,cell growth was not inhibited under test concentration.Colchicine andβ-cyclodextrin suitable for the growth of suspension cell concentrations were 50 mg·L-1 and 10 mg·L-1. In addition to salicylic acid inhibited the synthesis of resveratrol,the cinnamic acid, phenylalanine, colchicine andβ-cyclodextrin Conducive to the resveratrol synthesis of appropriate concentrations were 50 mg·L-1, 40 mg·L-1, 40 mg·L-1 and 40 mg·L-1.
