Plant male sterility is a common genetic phenomenon of nature, use of plant male sterile plants, hybrid seeds can get access to heterosis and improvement of plant disease resistance and reproductive performance. Therefore, crop research and application of male sterility has been the area of domestic and international hot spots of plant genetic breeding for decades by many scholars attention. Essentially reveal the mechanism of infertility, both from the scientific value, or in practice are of great significance.For the Photo-thermo-sensitive sterility maize importance, we conducted a preliminary study it with germination, planting of field trials, electrophoresis and MS.The long stored male sterility maintainer seeds germination experiments carried out through a series of experiments to select the optimal seed germination temperature and reagents, for a large number of male sterility maintainer seeds germination .Different Concentration of H202,H2SO4,NaOH and KNO3 were processed seeds. The results showed the effect of 95%H2SO4,30%NaOH and 30%H2O2better than the other goups. The effect of 30%H2O2 was the best on the rate of germination.Because male sterile plants in this experiment had four features: its tassel degradation, no flowers, no spike and white tassel, so the seed was required artificial pollination to preserved.Seeds were got through the pollination of maintainer, finally 18 lines maintainer were preserved.SOD,POD,CAT and soluble protein were tested in the four periods of male sterile line and maintainer (seedling stage, jointing stage, milky stage and mature stage).The results showed SOD firstly increased and then decreased, POD decreased totally, CAT grown in general, the soluble protein firstly increased and then decreased. This phenomenon indicated that the antioxidant capacity increased at first and then decreased in the four periods of maize.In the comparison of male sterile and maintainer of maize, the leaves SOD in male sterile maize was lower than it in maintainer. The stem SOD in male sterile maize was higher than it in maintainer. There was no clearly difference of root SOD between male sterile and maintainer of maize. The leaf POD in male sterile maize was lower than it in maintainer. The differences of the stem POD and the root POD respectively in male sterile and maintainer of maize were not obvious.CAT in leaf, stem and root of male sterile was lower than it in maintainer. The soluble protein of leaves and roots in male sterile was lower than it in maintainer, but it was reverse in the stem.SOD, POD and CAT was an important indicator of antioxidant ability in the plant, reflecting the ability of removing free oxygen, drought-resistance and disease-resistance. The value of stem and root was not obviously different, but the value of leave was different.In the SDS-PAGE, three group experiments were carried out to find the best condition.(1)3 methods of extracting leaves protein were compared.TCA/acetone was chose for its abundant bands and clear trial.(2) In the optimization of gel concentration, 15% gel was the best.(3) In sample quantity optimization, 20μL was the best.There were 5 different bands between the male sterile and maintainer of maize, among 14.4kD and 166kD. There was a new band in male sterile maize (No.1).Finally MS was employed to determine the differential protein(No.1).It may be annexin.annexin existed animals, plants and humans, a calcium-dependent phosphorylation of proteins. In maize, it may affect the membrane and Ca2+ binding, leading to abnormal development of cell organelles, and ultimately infertility. Or it may affect fertility protein gene expression, result in infertility.
