Male sterility of plants is a common genetic phenomenon in the nature. We can obtainplant hybrid seeds and heterosis by male sterility in plants,also we can enhance the plantdisease resistance and reproductive performance。Therefore, it is a research hotspot aboutMale Sterility Mechanism and application at home and abroad and many scholars focuson.there is a very important significance that reveal the mechanism of sterile from themolecular level for the genetic theory or the application practice. in order to illustrate themechanism of male sterile lines form physiology and molecular level, researching chloroplastprotein extraction,electrophoresis and mass spectrum of the Photo-thermo-sensitive sterilitymaize and fertile lineThere is significant difference of the chloroplast photosynthetic rate between Malesterile lines and the fertile line. Both of them reached the maximum at10to12pm. Thefertile line`s photosynthetic rate was generally higher.In order to study the chloroplast photosynthetic rate diffenece between Male sterilelines and the fertile line. researching chloroplast protein extraction.twice whet and extractmethod was build:grinding to a half broken, pour the liquid, adding extract grinding tocompletely broken.effusion chloroplast. primal separate with centrifugal force:3000g forserosity,Purification of centrifugal force:11150g.so holonomic chloroplast and protein canbe obtained.In the process of chloroplast protein extraction electrophoresis conditions wereoptimized:TCA/acetone extract protein,Gel concentration was15%,Sample volume was20μL. So differential protein were isolated in SDS electrophoresis. It shows that there isobvious different protein.Differentially expressed proteins were identified using Liquid Chromatography/MassSpectrometry. there are three proteins in this line.respectively are:1gi|22284,vicilin-likeembryo storage protein [Zea mays];2gi|242033447,hypothetical proteinSORBIDRAFT01g012640[Sorghum bicolor];3gi|222838014,predicted protein [Populustricho. carpa]。according to the mass spectrum result in the database. The first protein is storage protein similar to the pea protein which has a sensitization and may be associated withsterility.there is no report about others effect.maybe they are the reason maize male sterile,supplying information for maize male sterile mechanism further study.
