The author of this article separates 35 strains of fungi which can break down cellulose better from the forest humus soil and pig manure composting。Through screening of plate firstly and Re-screening of fermentation secondly,we get 5 strains of fungi which have the strong ability of enzyme production,named respectively:M-5, M-7, M-8, M-11, M-16,Of course, M-5 is the most of all.Through the Identification of morphology and ITS conserved sequences of rDNA, M-5 strain is identified as Aspergillus clavatonanicus.Research the solid-state fermentation conditions on M-5 strain,we can get the optimal conditions:carbon source are rice husk:bran=7:3, and the rice husk is pretreated by 3%NaOH;Nitrogen source are 1%(NH)2SO4;The ratio of solid to liquid 1:2.5; Inoculation volume of 105 spores/(g dry fermented matter) 1ml;PH 6.0,Cultured 84h around 30℃. Under the optimal conditions,CMC activity and FPA are 3015.4 umol/g.h and 432.9umol/g.h respectively.Studies on the character of CMC enzyme and filter paper enzyme of M-5 strain,we get the following results:optimal PH and temperature of CMCase are PH 8.0 and 50℃, filter paper enzyme PH5.0,50℃;We also study the effect of several metal ions on the enzyme activity; Finally,we study the stability of PH and temperature of CMC enzyme and filter paper enzyme respectively,they are both acidic and megatemperature enzymes,they both can withstand relatively high temperatures.M-5 strain as the main strain, other strains as bacteria-assisted,we study the role of microbial treatment of livestock and poultry manure for resource. The results shows that the inoculation of exogenous microbial preparation will help shorten the composting time, improve product quality composted;After the comparison of effect of composting among several microbial designs, It is found that design 3 has the best effect of composting; Compared to commonly used microbial preparation on the market, the effect of composting product of design 3 is little different, but the composting cycle is shorter and costs are significantly reduced, it will be a better microbial preparation for the use of composting.
