Research On The Variations Of Laccase Activity And Isozymes Of Two Toxicodendron Vernicifluum (Stokes) F.A. Barkley Varieties At Tapping Season In Shaanxi

Toxicodendron vernicifluum (Stokes) F. A. Barkley was an important kind of economical tree of Anacardiaceae Toxicodendron.Its main product was raw lacquer,which had a reputable name of "king of paint", was a kind of excellent natural paint and it was also the important material of modern technology, agriculture, national defense, civil and industry. Now the research in its structure, raw lacquer composition had been reported, but in laccase chemistry, the variations of laccase activity in Toxicodendron vernicifluum (Stokes) F. A. Barkley at tapping season had not been reported, and the research of laccae isozymes were more about epiphyte laccase and less in plant. In the prescent study, spectrophotometry was used to analyse the variations of laccase activity in Toxicodendron vernicifluum (Stokes) F. A. Barkley at tapping season, and four isozymes were successful segregated by isoelectrofocusing electrophoresis and mensurate the isoelectric point of each enzyme strap. The intention was providing reference for isozymes in Toxicodendron vernicifluum (Stokes) F. A. and the connection between isozymes and laccase activity, tapping time and variety of Toxicodendron vernicifluum (Stokes) F. A..1. The variations of laccase activity at tapping season embodies the same law between different varieties that laccase activity was decline form June to July, then rise from June to August, and lowest in June. But different variety had different characteristic.The hightest peak of laccase activity in Gaobachi laccase was in June, then Dahongpao laccase was in August. On the whole, the laccase activity in Gaobachi was higher than Dahongpao at tapping season.2. Molecular weight was mensurated by SDS-PAGE electrophoresis, which was 110 kDa. And there was other protein in extract.3. There were more than four isozymes in Toxicodendron vernicifluum (Stokes) F. A.by isoelectrofocusing electrophoresis, there isoelectric point were 6.85,6.98,7.16,7.35, which were clearly dyeing. Dahongpao lacquer enzyme electrophoresis results showed some difference with Gaobachi, there were no strap which isoelectric point was 6.85 in June, July and September but August. The relationship between laccase isozymes and the variations of laccase activity was further discussed.4. Establishing a method of rapid determination of enzyme isozyme which had stable, rapid, efficient advantages.
