Polymorphisms Of Sanjiang White Pigs GH And IGF-I Genes And Association Analysis With Parts Of Growth Traits

Abstract/Summary:
In this study, Sanjiang White pig population were taken as research object, the polymorphism of exon2 and 5 'terminal of GH gene in Sanjiang White pig were detected by PCR-RFLP method; and polymorphism of IGF-I gene in exon3 of Sanjiang White pig were detected by PCR-SSCP. And relationship between genotypes and growth traits were analyzed with GLM program of SAS (Version 9.0) statistical software. The results are as follows:1. Three mutation sites were detected in exon2 and 5 ' terminal of GH gene and in exon 3 of IGF-I gene in the Sanjiang White pigs, respectively. GH gene exon2 polymorphism site was controlled by pair of allele A and B, the mutation of A-G dominant were in 493bp,three genotypes including AA, AB, BB were detected, and the distribution tendency of these genotypes were AB>BB>AA; 5' terminal of GH gene polymorphism site was controlled by pair of allele C and D, the mutation of G-A dominant were in 154bp, three genotypes CC, CD, DD were detected, and the distribution tendency of these genotypes were CD>CC>DD;IGF-I gene exon 3 polymorphism site was resulted from C/T transition. It was controlled by pair of allel E and F. There were three genotypes, namely AA, AB, BB in IGF-I exon 3.Polymorphic information content (PIC) statistics showed that the three mutation sites were moderately polymorphism. The x2 test showed that the three mutations were in the Hardy - Weinberg equilibrium.2. The effects of different genotypes in GH gene exon 2 polymorphism site on growth traits was not significant (P>0.05), but the early growth traits of individuals with AA genotype was higher than that with AB and BB genotypes; the individuals with CC genotype and DD genotype had higher birth weight and weight to 60 days than the ones with CD genotype in 5' terminal of GH gene polymorphism site, but the difference was not significant(P>0.05).3. IGF-I gene exon3 polymorphism of different genotypes had clear effect on piglets birth weight(P<0.05). The birth weight of piglets with EE and FF genotype were higher than the ones with EF genotypes. The individuals with EE genotype had the highest weight to 60 days, followed by the ones with FF genotype, and the differences between three genotypes were significant (P<0.05).
